Oxytocin is required for normal milk fat production

ABSTRACT: We used single-nucleus RNA-seq to examine the heterogeneity of lactating mammary glands (MGs) and the transcriptional profiles of mammary epithelial cells (MECs) in control and adipocyte-specific oxytocin receptor knockout (OxtrΔAd) mice. Our analysis identified all expected MG cell types, including MECs, myoepithelial cells, adipocytes, adipose progenitors, hormone-sensing progenitors, vascular, and immune cells. Notably, OxtrΔAd MGs exhibited a profound metabolic shift in MECs, characterized by reduced mTOR signaling, increased autophagy, and decreased lipid synthesis. These findings underscore the essential role of OXT-driven adipose lipolysis in supporting lactation.
