Project description:Currently little is known about the safety of nonintoxicating cannabinoids such as cannabinol (CBN). New approach methodologies (NAMs) based on bioinformatic analysis of high-throughput transcriptomic data are gaining increasing importance in risk assessment and regulatory decision-making of data-poor chemicals. In this study, we conducted a concentration response transcriptomic analysis of CBN in hepatocytes derived from human induced pluripotent stem cells (iPSCs). Biological functions and pathways impacted by CBN exposure were identified, including liver metabolism and cellular oxidative stress. In addition, a transcriptomic point-of-departure (tPoD) value was determined for CBN through benchmark concentration (BMC) analysis of the transcriptomic data. The present study demonstrates the potential utility of transcriptomic BMC analysis as a NAM for hazard assessment of data-poor chemicals, improves our understanding of the possible health effects of CBN, and provides important tPoD data that could contribute to inform human safety assessment of cannabinoid compounds.

Project description:In the current study, we attempted to establish an in vitro model for assessing population variability in hepatotoxicity testing using a panel of hepatocytes derived from nine human induced pluripotent stem cell (iPSC) lines belonging to three different ethnic groups, Black or African American (BAA), Latino or Hispanic (LOH), and Non-Hispanic White (NHW). The panel of iPSC-derived hepatocytes were exposed to diglycolic acid (DGA) and usnic acid (UA), and transcriptomic changes were characterized and compared.

Gene expression data of hepatocytes derived from human fibroblasts, human iPSCs, and human adult hepatocytes

Project description:Generation of human fibroblast-derived hepatocytes capable of extensive proliferation, as evidenced by significant liver repopulation of mice. Unlike current protocols for deriving hepatocytes from human fibroblasts, ours did not generate iPSCs, but shortcut reprogramming to pluripotency to generate an induced multipotent progenitor cell (iMPC) stage from which endoderm progenitor cells (iMPC-EPCs) and subsequently hepatocytes (iMPC-Heps) could be efficiently differentiated. After transplantation into an immune-deficient mouse model of human liver failure, iMPC-Heps were able to engraft and proliferate, and acquired levels of hepatocyte function similar to adult hepatocytes. Microarray analysis has been used to show: 1. post-transplant maturation in vivo of iMPC-Heps compared to aHeps, 2. differences between iMPC-Heps and freshly isolated aHeps in vitro, 3. similar profile of freshly isolated aHeps to aHeps and iMPC-Heps in vivo, and 4. similarities of expression levels of most, but not all, genes between iMPC-Heps and iPSC-Heps in vitro. We isolated repopulating nodules of iMPC-Heps and aHeps by laser-capture microscopy (LCM) around 9 months after transplantation and analyzed their gene expression on the microarray, together with RNA from in vitro samples.

Project description:Organophosphorus compounds induce hepatotoxicity through currently unknown mechanisms, which need to be unraveled by a comprehensive and systematic approach such as genome-wide gene expression analysis. We used microarrays to study gene expression changes in human hepatocytes after exposure to VX, and identified pathways underlying these changes. Human hepatocytes were exposed to sublethal concentrations (0, 0.1, 10 μM) of VX. RNA were extracted at different timepoints (0, 6 h) after VX exposure and hybridized to Affymetrix microarrays. Four biological repeats were used for each condition.
