Effect of mechanical compression on gene expression in HEI-286 Schwann cells and pancreatic cancer cells MiaPaCa-2 and Panc01

ABSTRACT: In this study we compare transcriptional changes induced by mechanical compression in Schwann cells and pancreatic cancer cells. Human HEI-286 Schwann cells and pancreatic cancer cells MiaPaCa-2 and Panc01 were compressed at 5 μm height for 5 minutes in 6-wells static confiner (4Dcell) or uncompressed (not confined or confined in a 20 μm height space). Our data provides comparison of molecular response of Schwann cells and cancer cells to mechanical forces.

Project description:We report the transcriptome analysis of HEI-286 human Schwann cells and MiaPaCa-2 pancreatic cancer cells Nerves stimulate cancer progression for different cancer types, promoting both tumor growth and perineural invasion, a prominent process in pancreatic ductal adenocarcinoma. Yet little is known about how cells in the nerves contribute to cancer progression. Here we demonstrate that Schwann cells, the most abundant cell type in nerves, collectively function as tumor activated Schwann cell tracks (TASTs). Schwann cells in a 3D matrix organize into stellate tracks which serve as a conduit for cancer cell migration. TASTs actively promote pancreatic cancer dissemination during perineural invasion and neoneurogenesis. This key function of TASTs is induced by cancer cells triggering c-Jun activation of Schwann cells, analogous to the reprogramming that promotes nerve repair. In TASTs, dynamic Schwann cell processes wrap around and apply forces directly on cancer cells to enhance cancer motility. We define a model of cancer progression that relies upon collective Schwann cell behavior driven by c-Jun transcriptional reprogramming.

Project description:Loss of NF2 (merlin) has been suggested as a genetic cause of neurofibromatosis type 2 and malignant peripheral nerve sheath tumor (MPNST). Previously, we demonstrated that NF2 sustained TGF- receptor 2 (TR2) expression and reduction or loss of NF2 activated non-canonical TGF- signaling, which reduced RKIP expression via TR1 kinase activity. Here, we show that a selective RKIP inducer (novel chemical, Nf18001) inhibits tumor growth and promotes schwannoma cell differentiation into mature Schwann cells under NF2-deficient conditions. In addition, Nf18001 is not cytotoxic to cells expressing NF2 and is not disturb canonical TGF- signaling. Moreover, the novel chemical induces expression of SOX10, a marker of differentiated Schwann cells, and promotes nuclear export and degradation of SOX2, a stem cell factor. Treatment with Nf18001 inhibited tumor growth in an allograft model with mouse schwannoma cells. These results strongly suggest that selective RKIP inducers could be useful for the treatment of neurofibromatosis type 2 as well as NF2-deficient MPNST. To know the global effect of Nf18001, we performed the microarray with HEI-193.

Integration of transcriptomics and proteomics uncovers novel targets underlying the protective effects of Nrf2 knockout in HEI-OC1 cells

Project description:Cisplatin, a utilized anticancer drug in clinical practice, induces sensorineural hearing loss (SNHL) in patients. However, the precise mechanism underlying cisplatin-associated ototoxicity remains unknown. HEI-OC1 cells are immortalized cells derived from the organs of Corti mice and nuclear factor erythroid 2-related factor 2 （Nrf2） knockout (KO) significantly enhances cisplatin resistance in these cells. The exploration of transcriptomic data from Nrf2 KO has significant implications for the identification of novel targets to enhance HEI-OC1 cisplatin resistance in Nrf2 KO and for understanding the biological characteristics associated with SNHL. The RNA-seq analysis revealed a significant enrichment of differentially expressed genes (DEGs) in the Nrf2 KO model within key signaling pathways, including the PI3K-Akt, MAPK, as well as Glutathione metabolism signaling pathways. Notably, expression levels of 17 specific genes were confirmed by RT-qPCR (Real-time Quantitative-PCR). The biomarkers identified in this study may be key to understanding the biological mechanism by which Nrf2 KO strongly increases HEI-OC1 cisplatin resistance, and by targeting the PI3K-Akt, MAPK, Glutathione metabolism signaling pathways provide new ideas for the prevention and treatment of cisplatin-induced SNHL.
