The different genes expression in clinical para-tumor and pancreatic ductal adenocarcinoma samples

ABSTRACT: Pancreatic cancer is a highly malignant tumor. Current treatments are not effective. Hence, more effective treatment methods need to be sought. We aim to identify genes that differ between clinical para-tumor and pancreatic ductal adenocarcinoma samples by RNA-seq, and subsequently explore their functions, in the hope of finding better targets for pancreatic cancer treatment.

Project description:Pancreatic cancer is a highly malignant tumor that is well known for its poor prognosis. It has been reported that aspirin can reduce the risk of various cancers, but the potential role of aspirin and gemcitabine in pancreatic cancer pathogenesis and chemotherapy has not been studied. Therefore, we investigated their synergistic anti-tumor effects and explored the potential molecular mechanisms and biological functions underlying their inhibitory effects on the development of pancreatic cancer in the hope of identifying treatment-related or prognostic biomarkers for the effective treatment of pancreatic cancer.

Expression profiling of pancreatic ductal adenocarcinoma

Project description:Pancreatic ductal adenocarcinoma (PDAC) has the worst prognosis of all common cancers, but divergent outcomes are apparent between patients. To delineate the intertumor heterogeneity that contributes to this, we aimed to identify clinically distinct gene expression-based subgroups. From a cohort of 345 resected pancreatic cancer cases, 90 samples with confirmed diagnosis of PDAC and sufficient tumor content were available for gene expression analysis by RNA sequencing. Unsupervised classification was applied, and a classifier was constructed. Species-specific transcript analysis on matching patient-derived xenografts (PDX, N=14) allowed construction of tumor- and stroma-specific classifiers for use on PDX models and cell lines.

Dual inhibition of KRASG12D and pan-ERBB is synergistic in pancreatic ductal adenocarcinoma

Project description:Pancreatic ductal adenocarcinoma (PDAC) is a lethal cancer with a low survival rate. Recently, new drugs that target KRASG12D, a common mutation in PDAC, have been developed. We studied one of these compounds, MRTX1133, and found it was specific and effective at low nanomolar concentrations in patient-derived organoid models and cell lines harboring KRASG12D mutations. Treatment with MRTX1133 upregulated the expression and phosphorylation of EGFR and HER2, indicating that inhibition of ERBB signaling may potentiate MRTX1133 anti-tumor activity. Indeed, the irreversible pan-ERBB inhibitor, afatinib, potently synergized with MRTX1133 in vitro, and cancer cells with acquired resistance to MRTX1133 in vitro remained sensitive to this combination therapy. Finally, the combination of MRTX1133 and afatinib led to tumor regression and longer survival in orthotopic PDAC mouse models. These results suggest that dual inhibition of ERBB and KRAS signaling may be synergistic and circumvent the rapid development of acquired resistance in patients with KRAS mutant pancreatic cancer.

Whole-Tissue Gene Expression Study of Pancreatic Ductal Adenocarcinoma

Project description:Expression analysis of 36 pancreatic ductal adenocarcinoma tumors and matching normal pancreatic tissue samples from pancreatic cancer patients of the Clinical Institute Fundeni (ICF) using Affymetrix U133 Plus 2.0 whole-genome chips. Pairs of normal and tumor tissue samples were obtained at the time of surgery from resected pancreas of 36 pancreatic cancer patients. Gene expression was analyzed on Affymetrix U133 plus 2.0 whole genome microarrays. For three of the 36 normal-tumor sample pairs we carried out replicate microarray hybridizations in order to gauge the technical measurement errors. We thus performed 78 genechip hybridizations in total. A patient sample pair was excluded from further analysis since one of the samples did not meet the quality controls. The microarray data was subsequently normalized using the RMA algorithm.

Project description:Pancreatic ductal adenocarcinoma (PDAC) is one of the most lethal cancer due to its rapid progression, marked potential for metastasis and the difficulty in diagnosis1-3. However, there are no effective liquid tests currently available for PDAC detection besides CA19-9. Here we introduce a noninvasive detection approach that employs machine learning plus untargeted and targeted serum lipidomics to establish an accurate method to detect PDAC.

CRISPR genome-wide synthetic lethality screens of KRAS-driven mouse pancreatic ductal adenocarcinoma cells

Project description:We performed CRISPR genome-wide synthetic lethality screens to identify genetic vulnerabilities and chemogenetic interactions in KRAS-driven pancreatic cancer. We identify ERAD as a genetic target that synergizes with FTS treatment and we show that FTS treatment induces ER stress, which was further enhanced by inhibition of ERAD, leading to synergistic induction of apoptosis in pancreatic tumor cells. Altogether, our screens enable us to identify combination therapies for cancer treatment.
