Project description:The purpose of this study was to evaluate whether chronic, continuous hypoxia (breathing 11% FIO2) would improve neurodegenerative disease in a mouse model of Parkinson's disease, which was modeled through intrastriatal injection of a-synuclein preformed fibrils (PFFs). We find that chronic, continuous hypoxia helps to prevent further neurodegeneration and alleviate neurological disease.

Project description:Study objectives: Chronic obstructive pulmonary disease and obstructive sleep apnea overlap syndrome is associated with excess mortality, and outcomes are related to the degree of hypoxemia. People at high altitude are susceptible to periodic breathing, and hypoxia at altitude is associated with cardio-metabolic dysfunction. Hypoxemia in these scenarios may be described as superimposed sustained plus intermittent hypoxia, or overlap hypoxia (OH), the effects of which have not been investigated. We aimed to characterize the cardio-metabolic consequences of OH in mice. Methods: C57BL/6J mice were subjected to either sustained hypoxia (SH, FiO2=0.10), intermittent hypoxia (IH, FiO2=0.21 for 12 hours, and FiO2 oscillating between 0.21 and 0.06, 60 times/hour, for 12 hours), OH (FiO2=0.13 for 12 hours, and FiO2 oscillating between 0.13 and 0.06, 60 times/hour, for 12 hours), or room air (RA), n=8/group. Blood pressure and intraperitoneal glucose tolerance test were measured serially, and right ventricular systolic pressure (RVSP) was assessed. Results: Systolic blood pressure transiently increased in IH and OH relative to SH and RA. RVSP did not increase in IH, but increased in SH and OH by 52% (p<0.001) and 20% (p=0.001). Glucose disposal worsened in IH and improved in SH, with no change in OH. Serum LDL and VLDL increased in OH and SH, but not in IH. Hepatic oxidative stress increased in all hypoxic groups, with the highest increase in OH. Conclusions: Overlap hypoxia may represent a unique and deleterious cardio-metabolic stimulus, causing systemic and pulmonary hypertension, and without protective metabolic effects characteristic of sustained hypoxia.

Project description:Dissect the transcriptional consequences of neuroinflammation and the role of the kinase LRRK2 in regulating neuronal responses in human induced pluripotent stem cell (hiPSC)-derived neurons exposed to pro-inflammatory conditions relevant to Parkinson’s disease (PD) and Alzheimer’s disease (AD).Dopaminergic neurons (PD context) and cholinergic neurons (AD context) were differentiated from hiPSCs and subsequently cultured in glial-conditioned medium containing either α-synuclein pre-formed fibrils (α-syn pffs) or amyloid-β (Aβ) fibrils, for PD and aAD respectively. Neuronal cultures were exposed to inflammed media to model chronic inflammatory stress.

Project description:Lewy body inclusions enriched with aggregated forms of the presynaptic protein alpha-synuclein (aSyn) are a hallmark of synucleinopathy diseases such as Parkinson's disease (PD) and dementia with Lewy bodies (DLB). To model this pathology, C57 mice were injected in the cortex or striatum with preformed fibrils (PFFs) prepared from mouse aSyn to induce the aggregation of the endogenous mouse protein. Monomeric aSyn was injected as a control. Aggregated aSyn was detected via immunohistochemical staining in the sensorimotor cortex at 1- and 3-months post-injection in PFF-injected mice, but not monomer-injected animals. Phosphoproteomics analysis was carried out on homogenates from the sensorimotor cortex of mice inoculated with PFFs or monomer, 3 months post-injection, and the data are presented here.

Project description:Synucleinopathies are characterized by the accumulation and propagation of α-synuclein (α-syn) aggregates throughout the brain, leading to neuronal dysfunction and death. In this study, we used an unbiased FACS-based genome-wide CRISPR/Cas9 knockout screening to identify genes that regulate the entry and accumulation of α-syn preformed fibrils (PFFs) in cells. We identified key genes and pathways specifically implicated in α-syn PFFs intracellular accumulation, including heparan sulfate proteoglycans (HSPG) biosynthesis and Golgi trafficking. All confirmed hits affected heparan sulfate (HS), a post-translational modification known to act as a receptor for proteinaceous aggregates including α-syn and tau. Intriguingly, deletion of SLC39A9 and C3orf58 genes, encoding respectively a Golgi-localized exporter of Zn2+, and the Golgi-localized putative kinase DIPK2A, specifically impaired the uptake of α-syn PFFs, by preventing the binding of PFFs to the cell surface. Mass spectrometry-based analysis of HS chains in SLC39A9 -/- and C3orf58 -/- cells indicated major defects in HS maturation. Additionally, Golgi accumulation of NDST1, a prime HSPG biosynthetic enzyme, was detected in C3orf58 -/- cells. Interestingly, C3orf58 -/- human iPSC-derived microglia and dopaminergic neurons exhibited a strong reduction in their ability to internalize α-syn PFFs. Altogether, our data identifies new modulators of HSPGs that regulate α-syn PFFs cell surface binding and uptake.

Project description:Microglia, brain-resident immune cells, are involved in pathophysiology of several neurodegenerative diseases, including Parkinson’s disease. Given significant species-specific differences in microglia gene expression, particularly in disease-risk genes, as well as the highly reactive nature of these cells, studying human microglia in a whole brain environment is essential. Here, we established a humanized mouse model by transplanting human induced pluripotent stem cell-derived hematopoietic progenitor cells into the striatum of immunodeficient adult mice and injected human alpha-synuclein preformed fibrils to model Parkinson’s disease pathology. We aimed to create a novel model of Parkinson's disease and studied the response of the human microglia and mouse cells to the preformed fibrils.

Project description:Parkinson’s disease (PD) is a neurodegenerative disease characterized by progressive motor symptoms and alpha-synuclein (αsyn) aggregation in the nervous system. For unclear reasons, PD patients with certain GBA mutations (GBA-PD) have a more aggressive clinical progression. Two testable hypotheses that can potentially account for this phenomenon are that GBA1 mutations promote αsyn spread or drive the generation of highly pathogenic αsyn polymorphs (i.e., strains). We tested these hypotheses by treating homozygous GBA1 D409V knockin (KI) mice with human α-syn-preformed fibrils (PFFs) and treating wild-type mice (WT) with several αsyn-PFF polymorphs amplified from brain autopsy samples collected from patients with idiopathic PD and GBA-PD patients with either homozygous or heterozygous GBA1 mutations. Robust phosphorylated-αsyn (PSER129) positive pathology was observed at the injection site (i.e., the olfactory bulb granular layer) and throughout the brain six months following PFF injection. The PFF seeding efficiency and degree of spread were similar regardless of the mouse genotype or PFF polymorphs. We found that PFFs amplified from the human brain, regardless of patient genotype, were generally more effective seeders than wholly synthetic PFFs (i.e., non-amplified); however, PFF concentration differed between these two studies, and this might also account for the observed differences. To investigate whether the molecular composition of pathology differed between different seeding conditions, we permed Biotinylation by Antibody Recognition on PSER129 (BAR-PSER129). We found that for BAR-PSER129, the endogenous PSER129 pool dominated identified interactions, and thus, very few potential interactions were explicitly identified for seeded pathology. However, we found Dctn2 interaction was shared across all PFF conditions, and Nckap1 and Ap3b2 were unique to PFFs amplified from GBA-PD brains of heterozygous mutation carriers. In conclusion, both the genotype and αsyn strain had little effect on overall seeding efficacy and global PSER129-interactions.

Project description:The regulation of oxygen homeostasis is critical in physiology and disease pathogenesis. Studying transcriptomic profiles after hypoxia exposure could potentially uncover valuable biomarkers for predicting responses. To this end, 6 healthy individuals were recruited and exposed to graded normobaric hypoxia (FiO2 ~ 0.19 up till 0.13 in steps of 2% decrement in 10 minutes). A blood sample was collected before (at room air) and after the graded normobaric hypoxia exposure (at FiO2 ~ 0.13) for the gene-expression profiles. Results provide important information about early molecular differences induced by our experimental protocol in young, healthy individuals.
