CTNNB1 mutations have diverse effects on signalling in Embryonic Stem Cells

ABSTRACT: Bulk 3' RNA-Seq was performed on Embronic Stem Cells (E14) to explore the diverse effect of CTNNB1 mutation alongside GSK3β inhibition via CHIR99021 serving as a positive control.

Effect of activating Wnt signalling on expression of 3' UTR spliced transcripts in colorectal carcinoma cell line HCT116

Project description:The Wnt signalling pathway is commonly mutated or dysregulated in colorectal cancer, including approximately 93% of solid tumour samples from The Cancer Genome Atlas. Such dysregulation commonly leads to overactivation of CTNNB1, a transcription factor central to Wnt signalling. We have previously observed an increase in splicing of the 3'UTR of CTNNB1 in colon cancer samples compared to healthy samples. Additionally, we have observed isoform-specific regulation of CTNNB1 3'UTR spliced and retained isoforms in response culturing HCT116 cells with increasing concentrations of Wnt activator CHIR99021, and the opposite upon incubation with Wnt inhibitor IWR-1. By treating HCT116 cells with a high concentration of Wnt activator CHIR99021 and performing RNA sequencing, we assessed the global impact of Wnt signalling on expression of transcripts with spliced 3'UTRs. Interestingly, we observe an enrichment of alternative splicing events relating to components of the canonical Wnt signalling pathway itself, highlighting potential autoregulatory functions.

Transcriptomic analysis of gastric cancer cells in response to stable over-expression of circular RNA derived from CTNNB1 gene (circ-CTNNB1)

Project description:Circular RNAs (circRNAs), a subclass of noncoding RNAs characterized by covalently closed continuous loops, play emerging roles in tumorigenesis and aggressiveness. However, the functions and underlying mechanisms of circRNAs in regulating the Wnt/β-catenin signaling and cancer progression still remain elusive. We identify one intronic circRNA derived from CTNNB1 (circ-CTNNB1) as a novel driver of cancer progression. To investigate the mechanisms underlying the oncogenic functions of circ-CTNNB1, we employed the Illumina HiSeq X Ten as a discovery platform to analyze the transcriptome profiling changes of human gastric cancer AGS cells in response to stable over-expression of circ-CTNNB1. The results showed that stable over-expression of circ-CTNNB1 led to altered expression of 2230 human mRNAs, including 1244 up-regulated genes and 986 down-regulated genes. Furthermore, we validated the RNA-seq results by real-time RT-PCR with high identity. Overall, our results provided fundamental information about the transcriptomic changes in response to circ-CTNNB1 over-expression in human cancer cells, and these findings will help us understand the pathogenesis of cancer progression.

Project description:Mutations of β-catenin gene (CTNNB1) are frequent in adrenocortical adenomas (AA) and carcinomas (ACC). However, the target genes of CTNNB1 have not yet been identified in adrenocortical tumors. Our objective was to identify genes de-regulated in adrenocortical tumors harbouring CTNNB1 genetic alterations. We compared gene expression profiles of AA with (n: 3) and without (n: 4) CTNNB1 mutations using Affymetrix arrays.
