Scalable expansion and hepatic zone maturation of hepatic progenitor cells from human pluripotent stem cells

ABSTRACT: We devised a method for large-scale manufacturing of hepatocytes derived from human pluripotent stem cells by modulating levels of FGF19, TGFβ and NOTCH. We further employed WNT inhibitor and agonist to enhance differentiation efficiency. These differentiation strategies produce hepatocytes cells with improved in vivo functionality and enable their application in 3D bioprinted liver tissue constructs.
