Effect of adenoviral COVID-19 vaccines on human umbilical vein endothelial cells

ABSTRACT: Coronavirus disease 2019 (COVID-19) continues to cause outbreaks around the world despite the vast vaccination programs against it. Thus, it is important to keep developing better vaccines and other strategies to protect vulnerable populations. Here, we studied the safety of an adenovirus-based COVID-19 vaccine on human umbilical vein endothelial cells by total RNA sequencing and found, that the adenoviral vector vaccines had minimal effects on the gene expression profile of these cells.

Efficacy,safety and biodistribution of intranasally delivered adenoviral COVID-19 vaccines in hamsters

Project description:Coronavirus disease 2019 (COVID-19) continues to cause outbreaks around the world despite the vast COVID-19 vaccination programs. Thus, it is important to continue developing better vaccines to protect vulnerable populations. Here, we studied the efficacy, safety, persistence and biodistribution of an adenovirus-based COVID-19 vaccine delivered intranasally. Our results confirmed that mucosal delivery leads to effective protection against infection in hamsters. A comparative study of different adenoviral vectors demonstrated that specifically Ad5-S-3M, expressing the SARS-CoV-2 spike glycoprotein ectodomain with inclusion of three common receptor binding domain mutations, induced a robust humoral immune response and effectively protected hamsters from SARS-CoV-2 infection. In addition, longitudinal analysis of vaccinated hamsters’ tissues demonstrated durable vaccine transgene expression and persistent vector DNA in the nasal turbinates. This persistence may be beneficial in increasing the duration of immunity. Expression of the S-3M did not cause any aberrant changes in the hamster tissues and resulted in very few changes in cellular mRNA expression in target tissues. Safety and biodistribution assessment demonstrated restricted y tissue distribution of intranasally delivered vector and no significant changes in clinical blood chemistry, underlining the safety of the vaccine. These results further support that intranasal delivery is a promising approach for mucosal vaccine administration against respiratory infection pathogens and extend our knowledge of Adeno-vector persistence.

Project description:Functional T cell responses are crucial for protective immunity induced by COVID-19 vaccination, but factors influencing the quality of these responses are incompletely understood. We employ an activation induced marker (AIM) assay and single-cell transcriptomic sequencing to analyze SARS-CoV-2 spike-responsive T cells following mild SARS-CoV-2 infection or following one or two doses of mRNA-LNP or adenoviral vectored COVID-19 vaccines. Our findings reveal broad functional and clonal heterogeneity in T cells generated by vaccination or infection, including multiple distinct effector populations. T cell function was largely conserved between COVID-19 vaccine platforms but was distinct compared to SARS-CoV-2 infection. Notably, the dosing interval greatly influenced the quality of T cells after two vaccine doses, particularly after mRNA-LNP vaccination, where a longer interval led to reduced inflammatory signaling and increased secondary proliferation. These insights enhance our understanding of SARS-CoV-2 specific T cells and inform the optimization of mRNA vaccination remens.

Project description:Vector-based SARS-CoV-2 vaccines such as AstraZeneca's ChAdOx1 nCoV-19 have been associated with vaccine-induced thrombosis (TTS/VITT). We studied ChAdOx1 nCov-19 (AstraZeneca) and Ad26.COV2.S (Johnson & Johnson) and compared the vaccines using proteomics with mass spectrometric measurements. We could demonstrate that ChAdOx1 nCoV-19 contained significant amounts of human proteins in addition to adenoviral proteins. This was not the case for Ad26.COV2.S. Among the detected human proteins, proteasome subunits beta 1/delta and beta 5/MB1 were identified. In summary, both studied SARS-CoV-2 vaccines are markedly different concerning their content of contaminating proteins, which is most likely a determining factor for different frequencies of TTS/VITT after their application.

Intranasal administration of adenoviral vaccines expressing SARS-CoV-2 spike protein improves vaccine immunity in mouse models

Project description:Intranasal vaccines can prime or recruit to the respiratory epithelium mucosal immune cells capable of preventing transmission of SARS-CoV-2. We found that a single intranasal dose of serotype 5-based adenoviral vectors expressing either the receptor binding domain (Ad5-RBD) or the complete ectodomain (Ad5-S) of the SARS-CoV-2 spike protein was effective in inducing i) secretory and serum anti-spike IgA and IgG, ii) robust SARS-CoV-2-neutralizing activity in the serum and in respiratory secretions, iii) rigorous spike-directed T helper 1 cell/cytotoxic T cell immunity, and iv) protection of wild-type mice from a challenge with the SARS-CoV-2 beta variant. Our data confirm and extend previous studies reporting promising preclinical results on vector-based intranasal SARS-CoV-2 vaccination, and support the potential of this approach to elicit mucosal immunity for preventing reinfection and transmission of SARS-CoV-2 more effectively than the currently available vaccines.

Time-dependent enhancement of mRNA vaccines by 4-1BB costimulatory antibodies

Project description:mRNA vaccines have demonstrated efficacy against COVID-19. However, concerns regarding waning immunity and breakthrough infections have motivated the development of next-generation vaccines with enhanced efficacy. In this study, we investigated the impact of 4-1BB costimulation on immune responses elicited by mRNA vaccines in mice. We first vaccinated mice with an mRNA vaccine encoding the SARS-CoV-2 spike antigen like the Moderna and Pfizer-BioNTech vaccines, followed by administration of 4-1BB costimulatory antibodies at various times post-vaccination. Administering 4-1BB costimulatory antibodies during the priming phase did not enhance immune responses. However, administering 4-1BB costimulatory antibodies after 96 hours elicited a significant improvement in CD8 T cell responses, leading to enhanced protection against breakthrough infections. A similar improvement in immune responses was observed with multiple mRNA vaccines, including vaccines against common cold coronavirus, human immunodeficiency virus (HIV), and arenavirus. These findings demonstrate a time-dependent effect by 4-1BB costimulation and provide insights for developing improved mRNA vaccines.

Different COVID-19 vaccines induce distinct reprogramming of stromal functions in draining lymph nodes

Project description:The non-leukocytic stromal cells of lymph nodes critically regulate immune responsiveness. However, the effects of different SARS-CoV-2 vaccines on stromal cell biology are unknown. We used single-cell transcriptomics to study early responses of stromal cells in draining lymph nodes after immunizing mice with clinically used COVID-19 vaccines, namely Spikevax®, Comirnaty®, Vaxzevria® and Nuvaxovid™. We found that vaccinations lead to robust transcriptomic changes, including vaccine-selective ones, in the different lymph node stromal cell populations priming the lymph node for the upcoming adaptive immune response.
