Project description:Drought is a harsh abiotic stress, with plants possessing diverse strategies to survive periods of limited water resources. Although previous research has established links between strigolactone (SL) and drought, in this study, we used the barley (Hordeum vulgare) SL-insensitive mutant hvd14 (dwarf14) to scrutinize the SL-dependent mechanisms associated with water deficit response. We have employed a comprehensive approach integrating transcriptome, proteome, phytohormone analyses, and physiological data to unravel differences between wild-type and hvd14 plants when responding to drought.

Differential gene expression analysis of drought responsive sense and antisense genes in Populus

Project description:In this study we employ a strand-specific RNA-seq appoach and stranded gene expression analysis tools to identify drought responsive antisense gene loci and sense-antisense gene pairs in Populus. we generated and sequenced 28 strand-specific cDNA libraries derived from either leaf or root tissues of Populus trichocarpa plants associaed with both short-term drought (24 hours of water stress of 40% of field capacity) and long-term drought ( 25 days of water stress of 40% of field capacity) . We mapped over 71 billion nucleotides to Populus genome. Our data demonstrates that with the current sequence depth ~ 19 % of Populus genome undergoes antisense transcription subjected to drought regulation. All in all we have identified that in root tissues 524 differentially expressed antisense genes and 247 drought-responsive SA gene pairs which are significantly regulated by drought (padj <0.05). Taken all data from both drought treatments, we have identified 1185 unique drought-responsive antisense gene loci and 606 drought-responsive SA gene pairs (padj <0.05).

Uncovering the positive role of strigolactone perception in barley drought response I

Project description:Drought is a destructive abiotic stress, with plants possessing diverse strategies to survive periods of limited water resources. Previous results have described connections between strigolactone (SL) and drought, however, here we used the barley (Hordeum vulgare) SL-insensitive mutant hvd14 (dwarf14) to investigate the SL-dependent mechanisms related to water deficit response. By combining transcriptome, proteome with phytohormone analyses and physiological data, we describe the drought-mediated differences between wild-type and hvd14 plants. Our findings indicate that the drought sensitivity of hvd14 is related to weaker induction of abscisic acid-responsive genes/proteins, lower jasmonic acid content, higher reactive oxygen species content, and lower wax biosynthic and deposition mechanisms then wild-type plants. In addition, we identify a series of transcription factors (TFs) that are exclusively drought-induced in wild-type barley. Critically, we resolve a comprehensive series interestions between the drought-induced barley transcriptome and proteome responses that allow us to understand the impacts of SL in mitigating water limiting conditions. These data provide a number of new angles for the development of drought-resistant barley.

Mapping drought-responsive genes in cotton

Project description:Structural and functional approaches were used to study cotton (Gossypium) genes implicated in water-deficit stress. A genetic map representing the hypothetical ancestral diploid genome (Consensus Map) was used to map 1,907 of 15,784 tentative consensus sequences (TCs). These TCs represent 25,119 cotton ESTs derived from various tissues under irrigated and water-limited condition. The correspondence of mapped TCs and 42 stress-related quantitative trait loci (QTLs) revealed that 391 of the initial 1907 TCs co-localized within a QTL interval. About 31% of these TCs were annotated as genes involved in plant responses to abiotic stress. By comparison, only 18% of the total annotated TCs mapped on the Consensus map were classified as abiotic stress genes. The enrichment of stress-related TCs that map to stress-related QTLs could not be explained by chance (P = 1.5 x 10-7). Gene expression profiling experiments were carried out using a microarray composed of 12,006 oligonucleotides. Transcriptional responses to imposed water-deficit stress in root and leaf tissue of 8-week old cotton plants revealed 1401 transcripts identified as drought responsive. A total of 158 (84 drought-induced and 74 drought-repressed) genes were mapped, of which 22 (8 induced and 14 repressed) genes co-localized with a QTL. A total of 539 unique genes were identified in the drought-stressed libraries. However, only 91 of these genes are contained on the array. Of these genes, 12 showed significant changes in transcript abundance between stressed and irrigated leaf and root. Forty-five candidate genes implicated in drought-stress response at some level of characterization were identified. Keywords: stress response
