Effects of deleting Alg14, Hex1, Hex2, GPI1, or Hxk1 on changes of transcriptomic profiles in Beauveria bassiana


ABSTRACT: Beauveria bassiana is a popular and eco-friendly biopesticide. N-acetylglucosamine (GlcNAc) is a building block of both insect exoskeleton and fungal cell wall. GlcNAc-metabolic related genes might play vital roles during B. bassiana pathogen-pest interaction. Previous studies in Candida albicans have demonstrated that a UDP-GlcNAc transferase unit (Alg14), beta-hexosaminidase (Hex1 or Hex2), N-acetylglucosaminyl transferase component (GPI1), and hexokinase (Hxk1) was involved in GlcNAc-related metabolism. Herein, we found each homology of Alg14, Hex1 or 2, GPI1, and Hxk1 in B. bassiana, and reported the alteration of transcriptomic profiles between wild-type strain and gene-deletion mutants grown on cuticle broth (CB) supplemented with 1% cuticle powder of Locusta migratoria exoskeleton, followed by shaking at 150 rpm for 5 days at 25 degree centigrade.
