Biomarkers for Predicting Therapeutic Response to Preoperative Chemoradiotherapy in Rectal Cancer

ABSTRACT: This study analyzed 102 rectal cancer patients treated with PCRT and surgery to identify molecular subtypes predicting treatment response. Using transcriptomic data and pathway-based clustering, an immunogenic cluster was identified, showing high immune activity and a 90.3% favorable response rate. The findings were validated in external datasets, and a 16-gene predictive model demonstrated high accuracy. These results suggest that immune-related gene expression can guide personalized treatment in rectal cancer.
