RNA sequencing analysis of photon irradiated 5-, 7- and 11-day old thyroid gland organoids

ABSTRACT: The specific effects of radiotherapy treatment on thyroid physiology remain poorly understood. In our previous work, we characterized thyroid gland organoids grown from isolated mice thyroid cells. Herein, we used this model to investigate the events occurring at 2 days post-irradiation (corresponding to 7-day-old organoids) and at 6 days post-irradiation (corresponding to 11-day-old organoids) using bulk RNA sequencing. To compare the expression profile to an healthy standard sample, 5-day-old organoids (day of irradiation) were used as controls. This analysis outlined key dysregulated pathways activated in thyroid cells with photon exposure.

Project description:The specific effects of different radiation types, such as photons and protons, on normal stem cell activity and tissue regeneration remain poorly understood. In our previous work, we characterized salivary gland organoids at 2 days post-irradiation (corresponding to 7-day-old organoids) using single-cell RNA sequencing. To compare the cellular composition of 5-day-old organoids (day of irradiation) with that of 7-day-old organoids, we performed bulk RNA sequencing and applied the deconvolution tool CIBERSORTx, using the 7-day single-cell dataset as a reference. This analysis confirmed that the cell populations in 5- and 7-day-old salivary gland organoids were comparable.

Project description:The aim of our study is to investigate and compare the effects of carbon and photon irradiation on microvascular endothelial cells. Therefore we irradiated human pulmonary microvascular endothelial cells (HPMEC) with either 2Gy Carbon or 6Gy Photon (bioequivalent doses) and performed microarray analysis both 2 hours (short-term effect) and 6 days (long-term effects) after irradiation. All experiments were performed in 3 biological replicates.

Transcriptional responses in various normal mouse tissues after 24 h following 4 MV photon beam irradiation using three partial body irradiation set-ups

Project description:Female BALB/c nude mice (n=3/group) were subjected to partial body irradiation under anesthesia using 2 Gy external photon (4 MV, nominal) irradiation. In group A, the collum (i.e. the thyroid) was irradiated; in group B, thorax+abdomen were irradiated, and in group C, collum+thorax+abdomen were irradiated. The control group (n=5) was anesthetized but not irradiated. At 24 h after treatment, the kidneys, liver, lungs, spleen, and thyroid were excised, flash-frozen, and stored at -80°C. Total RNA was extracted from homogenized tissue samples (kidney cortex and kidney medulla were treated separately) and subjected to expression analysis using RNA microarray technology.

Project description:The aim of our study is to investigate the effects of carbon ion and photon irradiation on A549 tumor cells and analyse how these effects are altered by PML knockdown. Therefore we created PML knockdown A549 cells (shPML) and irradiated them with either 2Gy carbon ion or 6Gy Photon (bioequivalent doses). 4 days after irradiation microarray analysis was performed. All experiments were performed in 3 biological replicates and control groups were transduced with an empty vector.

Transcription profiling of prostate tumor cell line AT1 inocculated into rats and then subjected to carbon ion or photon radiation

Project description:In the present study approximately 1 to 2 mm3 prostate tumor AT1 was inoculated subcutaneously in the right hind leg of adult male Copenhagen rats. When the tumor diameter exceeded 15 mm, tumors of 5 and 4 rats were irradiated with carbon ion radiation of 37 or 16Gy respectively. Tumors of 5 other rats were irradiated with photon radiation of 37Gy. One animal irradiated with 37 Gy carbon ion radiation and one animal irradiated with photon radiation was sacrificed 12h, 30h, 72h, 7d and 14d after irradiation respectively. One animal irradiated with 16 Gy photon radiation was sacrificed 12h, 60h, 7d and 14d after irradiation respectively. Non-irradiated animals were sacrificed at 60h time point. Tumors were dissected and frozen in liquid nitrogen immediately. Total RNA from tumor material was isolated using the NucleoSpin RNA L kit (#740962.20, Macherey-Nagel). Differential gene expression analysis was performed on the Agilent whole rat genome Oligo Microarray (44k) platform by comparative two dye hybridisation with dye-swaps.
