RNAseq analysis of bone marrow plasma cells from Taciflox/flox and Jchain-CreERT2;Taciflox/flox mice

ABSTRACT: Durable neutralizing antibodies are essential for vaccine-induced protection and are continuously secreted by bone marrow long-lived plasma cells (LLPCs). The longevity of these cells largely dictates the duration of vaccine-induced immune protection. We perfomred conditional deletion of TACI in PCs to analyze the effect of TACI on bone marow plasma cellls using Jchain-CreERT2;Taciflox/flox mice.

To investigate a previously unappreciated role of splenic LLPCs, we sorted splenic plasma cells (PCs) from K/BxNsf and K/BxN mice, corresponding to LLPCs and conventional short-lived PCs, respectively, and compared their transcriptomes.

Project description:Newly generated plasma cells in secondary lymphoid organs migrate to niches in the bone marrow, wherein they survive as long-lived plasma cells (LLPCs). Although characterization of LLPCs has been well performed, it is still unclear what is key determinant(s) for longevity of plasma cells. One postulated model is that heterogeneity of plasma cells is formed by such as BCR affinity and/or microenvironment at the induction site, and some of these variables may instruct their longevity. Here, we found that, among newly generated IgG plasma cells, integrin b7hi marks plasma cells prone to home to the bone marrow, whereas integrin b7lo cells remain in secondary lymphoid organs.

Transcriptome analysis of TACI-deficient and WT naïve T cells

Project description:TACI (transmembrane activator and calcium modulator and cyclophilin ligand interactor) plays critical roles in B cells by promoting immunoglobulin class-switching and plasma cell survival. However, its expression and function in T cells remain controversial. In this study, we found that TACI deficiency in mice results in expansion of TH17 and Treg populations. To understand the mechanisms underlying increased frequencies and numbers of TH17 cells correlated with more severe colitis in TACI-/- mice, microarray-based transcriptome analysis was conducted to identify genes that were differentially expressed in TACI-/- vs WT naive CD4+ T cells.

Microarray for B cells of TACI-IgG-treated MRL/lpr mice compared to control mice

Project description:Both multiple myeloma (MM) and systemic lupus erythematosus (SLE) are characterized with abnormal production of plasma cells. In both diseases, the process of B cells differentiate into plasmablast/plasma cell is disordered. Despite the continuous research on the development of prognostic factors and introduction of new agents, dysregulation of plasmablast/plasma cells in MM and SLE is still uncontrolled. Thus, it is necessary to explore the novel therapeutic target to plasmablast/plasma cells. We and other researchers have shown that BAFF inhibitor atacicept (TACI-IgG), leads to some degree of B cell depletion. BAFF-specific targeted therapy specifically affects early-stage B cells in the periphery without affecting late-stage compartments such as plasma cells. Specifically depletion of plasma cells could hold great potential for the treatment of autoimmune diseases. To explore the novel therapeutic target to plasma cells, we determined the gene expression profile in B cells (mainly plasma cells) from atacicept (TACI-IgG)-treated lupus-prone MRL/lpr mice by affymetrix microarrays.

Transcriptome of TACI KO and WT mouse marginal zone and follicular B cells

Project description:The mature peripheral B cell compartment consists of follicular (FO) and marginal zone (MZ) B cells, which mature from transitional B cells in the spleen and mount T-dependent and T-independent antibody responses, respectively. TACI (Tnfrsf13b) is a member of the TNF-receptor superfamily expressed on mature and transitional B cells, with highest expression on MZ B cells and plasma cells, which is activated by the cytokines BAFF and APRIL. BAFF also stimulates the related receptor BAFFR, which provides signals that are essential for mature B-cell survival. Previous studies of TACI-/- mice have observed that TACI-deficiency leads to an expansion of mature B cells, and therefore it has long been thought that TACI is a negative regulator of mature B-cell survival. However, TACI-/- mice also have elevated levels of BAFF, therefore the B-cell hyperplasia phenotype in TACI-/- mice is most likely a cell-extrinsic effect due to increased BAFF-BAFFR signalling in TACI-/- B cells. To investigate the cell-intrinsic role of TACI, we generated mouse mixed bone marrow chimeras from TACI-/- and WT mice and studied the effect of TACI loss in vivo, without the confounding factor of increased BAFF. Here, we have analysed the transcriptomes of TACI KO and TACI WT marginal zone and follicular B cells by RNAseq.

Myeloid Skewing in Aging Bone Marrow Depends on Long-lived Plasma Cell Accumulation

Project description:Aging results in a reduction in B lymphopoiesis and an increase in myelopoiesis in mice and humans. We investigated how cellular changes in the aging hematopoietic microenvironment contribute to these alterations in hematopoiesis and found that long-lived plasma cells (LLPCs) increase in number in the bone marrow of old mice. The LLPCs exhibited a Toll Like Receptor activation signal, and their accumulation was partially dependent on inflammatory cytokines. LLPCs were able to stimulate myelopoiesis from hematopoietic stem cells and myeloid progenitors in vitro, and anti-CD138 antibody-mediated depletion of LLPCs or blockade of pro-inflammatory signaling in old mice in vivo resulted in a significant reduction in LLPC number and attenuated myeloid development. However, B lymphopoiesis remained suppressed. These data identify the increase in LLPC number as obligate for age-associated increases in myelopoiesis and provide evidence that, in the context of aging, enhanced myelopoiesis and depressed lymphopoiesis are distinct, independently regulated processes.

Gene expression profile at single cell level of rotavirus specific CD4+ T cells

Project description:IgA is the dominant antibody isotype secreted into the mucosal lumen. Mucosa-resident IgA+ plasma cells (PCs) generated following enteric infection can persist as long-lived plasma cells (LLPCs) in the gut, ensuring durable protection against reinfection. However, little is known about how virus-specific IgA+ PCs are generated and maintained. Upon infection with mouse rotavirus (RV) via the oral route, we found that antigen (Ag)-specific IgG+ and IgA+ PCs are generated in a follicular helper T (TFH) cell-dependent manner that involves CD40 signaling and MHC class II (MHCII) restricted Ag-presentation by B cells. Unexpectedly, although the RV-specific IgG response requires Ag-presentation by conventional dendritic cells, Ag-presentation by B cells is sufficient for the generation of RV-specific IgA. The accumulation of RV-specific IgA+ PCs in the small intestinal lamina propria (SILP) was found to depend on a type 1 TFH (TFH1) cell subset, which requires B cell Ag-presentation for their development and co-expresses Bcl-6 and T-bet. IFNγ receptor signaling is required for the expression of CXCR3 on RV-specific IgA+ PCs, which in turn promotes their migration to the SILP. Moreover, TFH1 cells are also required for the optimal development of IgA+ LLPCs in the upper and lower respiratory tracts in response to intranasal influenza A virus (IAV) infection. However unlike with RV, expression of MHCII on B cells was not sufficient for generating an IAV-specific IgA response in the airways, suggesting the operation of mucosal site-specific priming mechanisms. Collectively, our data reveal that unconventionally primed TFH1 cells support IgA responses to mucosal viral infections.
