Integrated Transcriptome and Metabolome Analysis Reveals Novel Metabolic Pathways and Metabolites in Moderate-to-Severe Psoriasis


ABSTRACT: Background:Psoriasis is a common immune-mediated disease; however, the understanding of its metabolic pathways remains incomplete. Methods:Blood samples from 11 patients with psoriasis and 11 matched healthy controls were subjected to integrated transcriptomic and untargeted metabolomic profiling, followed by comprehensive statistical analysis.Results:We identified 4,887 metabolites and 53 metabolic pathways. Integration with transcriptomic data from the patients revealed six co-regulated pathways: arachidonic acid, central carbon metabolism in cancer, cysteine and methionine, lysine degradation, propanoate, and tryptophan metabolism. Further analysis revealed associated gene and metabolite alterations. To our knowledge, central carbon metabolism in cancer is newly linked to psoriasis. The other pathways are poorly documented; our results validate and extend previous reports.Conclusions:Integrated multi-omics profiling identified six dysregulated metabolic pathways in psoriasis, including a novel association with central carbon metabolism in cancer. Our findings confirm and extend knowledge on poorly documented pathways, offering new insights into psoriasis pathogenesis and potential therapeutic targets.
