MBD3 regulates thymic Treg precursor cell development by modulating CD25 expression [ATAC-seq]

ABSTRACT: ATAC-seq was performed on thymic T cell subsets, focusing on immature CD4 single-positive (ImCD4SP) cells, as well as pre-tTreg and mature tTreg cells, from wild-type (WT) and Mbd3 conditional knockout (CKO) mice. Chromatin accessibility changes were most pronounced in the ImCD4SP stage, consistent with RNA-seq data indicating that transcriptional differences arise early, likely reflecting altered responses to stimulation preceding pre-tTreg specification. Loss of MBD3 altered accessibility at regulatory regions of Treg-associated genes. These ATAC-seq datasets offer insight into the epigenetic mechanisms by which MBD3 shapes early thymic Treg development.
