Tumour endothelial cells transcriptomics in response to Doxorubicin treatment.

ABSTRACT: Despite its established role in breast cancer treatment, Doxorubicin remains a challenge in breast cancer treatment as its long-term success is prevented by adaptive resistance mechanisms that extend beyond cancer cell intrinsic alterations. Our study in a MMTV-PyMT-driven mouse breast cancer model reveals that prolonged Doxorubicin exposure triggers a significant reprogramming of the tumour vasculature, substantially altering the angiocrine landscape and shaping treatment outcomes.

Project description:Phosphoproteomic analysis was performed to decipher changes occurring in tumour cells after treatment with conditioned media from endothelial cells, which may protect them from doxorubicin damage, and promote resistance to the treatment. Briefly, MMTV-PyMT (mouse mammary tumour virus-polyoma middle tumour-antigen) derived tumour cells were pretreated with conditional media from endothelial cells isolated from MMTV tumours treated with doxorubicin for 24 hours, andthen treated afterwards with vehicle (PBS) or doxorubicin in culture for 45 minutes and 2 days.

Protein difference of breast cancer tissue in mice after radiation therapy and without radiation

Project description:In this study, radiation therapy was performed on mouse breast cancer spontaneous gene mice (MMTV-PyMT) to obtain radiation therapy model of breast cancer mice. To simulate clinical breast cancer radiotherapy and study the differential proteins of breast cancer radiotherapy resistance. MMTV-PyMT mice were randomly divided into two groups, and the radiotherapy group was given the same dose 5 times (3GY, once every 2 days) when the breast cancer tumor grew to an appropriate size (200mm3). The protein extracted from mouse tumor was analyzed by mass spectrometry.

Project description:The tyrosine kinase ErbB2 positive breast tumors have more aggressive tumor growth, poorer clinical outcome, and more resistance to radiotherapy, chemotherapy and hormone therapy. To identify the biomarkers for predicting the responses to chemotherapy, we treated genetically diverse F1 backcross (F1Bx) mice between C57BL/6J and FVB/N MMTV-Erbb2 transgenic mice with mammary tumor by doxorubicin. Biopsies were taken in sterile conditions and processed for RNA and histological analysis before treatment. This will help us to have a better understanding of the heterogeneous response to chemotherapy and permit us to design better and more individualized (personalized) treatment strategies for human ErbB2 positive breast cancer.
