RNA-seq analysis of PAPD5 inhibitor effects in expanded human induced pluripotent stem cell-derived cardiomyocytes
Ontology highlight
ABSTRACT: Human induced pluripotent stem cells (hiPSCs) were differentiated into cardiomyocytes using a conventional monolayer protocol to generate non-expanded cardiomyocytes (N-CMs). A subset of cardiomyocytes was further replated and cultured in the presence of CHIR99021 to generate expanded cardiomyocytes (E-CMs). During the expansion phase, E-CMs were treated with the PAPD5 inhibitor BCH001 (E-CM-B) or RG7834 (E-CM-R) to prevent cellular senescence. RNA sequencing was performed to characterize transcriptional differences associated with senescence and mitochondrial maturation. Treatment with BCH001 during cardiomyocyte expansion attenuated the expression of senescence-associated markers and promoted gene expression profiles consistent with mitochondrial maturation. This dataset provides transcriptomic insights into the molecular effects of PAPD5 inhibition on expanded hiPSC-derived cardiomyocytes.
