Application of Type XVII Collagen in the Repair and Healing of Skin Defects

ABSTRACT: This work elucidates the crucial role of type XVII collagen in cutaneous wound healing and discusses the potential mechanisms through which it promotes tissue repair, including modulation of inflammation, enhancement of keratinocyte migration, regulation of ROS homeostasis, and support of basement membrane integrity.

Project description:Type XVII collagen (COL17A1) is well-characterized as a critical structural component of the epidermal basement membrane in the skin; however, its presence and function in the urinary tract remain poorly described. We discovered that COL17A1 is also expressed in the urothelium under urological condition. To elucidate the molecular mechanisms underlying early urothelial abnormalities caused by COL17A1 deficiency, we performed transcriptomic profiling (bulk RNA-seq) of ureter tissues from 2-week-old wild-type and Col17a1 knockout mice.

Project description:We searched for candidate genes responsible for Epithelial-to-mesenchymal transition (EMT) by microarray analysis of cultured lung cancer cells with or without the property of spheroid formation. We found increased expression of a variety of adhesion molecules in cancer stem-like cells, including Collagen XVII. Further functional assays and signaling pathway were also investigated.

Gut microbiota-induced perturbations in bile acids alter keratinocyte lipid metabolism via FXR-NQO1 signaling in psoriasis

Project description:Emerging evidence has shown that microbiota dysbiosis and aberrant bile acids (BAs) profiles are correlated with disease progression. This study elucidates dysregulated BAs metabolism in psoriasis patients and imiquimod-treated female mice, coupled with increased expression of the farnesoid X receptor (FXR) in keratinocytes. Activation of FXR by glycochenodeoxycholic acid (GCDCA) ameliorates psoriatic symptoms by enhancing lipid metabolism, particularly fatty acid oxidation. Mechanistically, the FXR-mediated enhancement of antioxidant capacity by upregulating NAD(P)H quinone dehydrogenase 1 (NQO1) expression underlies its regulatory role in lipid metabolism, offering an insight into FXR’s role in oxidative stress and lipid metabolism. Conversely, keratinocyte-specific FXR ablation exacerbates psoriasis severity. Gut microbiota dysbiosis is further identified as a pivotal contributor to perturbations in BA profiles in psoriasis. These findings support a mechanistic model linking gut microbiota and BA alterations to FXR signaling in keratinocytes and psoriasis-associated metabolic dysregulation, suggesting therapeutic potential for microbiota-targeted interventions.
