Rapamycin/sodium hyaluronate binding on nano-hydroxyapatite coated titanium surface improves MC3T3-E1 osteogenesis.

Project description:Endosseous titanium (Ti) implant failure due to poor biocompatibility of implant surface remains a major problem for osseointegration. Improving the topography of Ti surface may enhance osseointegration, however, the mechanism remains unknown. To investigate the effect of modified Ti surface on osteogenesis, we loaded rapamycin (RA) onto nano-hydroxyapatite (HAp) coated Ti surface which was acid-etched, alkali-heated and HAp coated sequentially. Sodium hyaluronate (SH) was employed as an intermediate layer for the load of RA, and a steady release rate of RA was maintained. Cell vitality of MC3T3-E1 was assessed by MTT. Osteogenesis of MC3T3-E1 on this modified Ti surface was evaluated by alkaline phosphatase (ALP) activity, mineralization and related osteogenesis genes osteocalcin (OCN), osteopontin (OPN), Collagen-I and Runx2. The result revealed that RA/SH-loaded nano-HAp Ti surface was innocent for cell vitality and even more beneficial for cell osteogenesis in vitro. Furthermore, osteogenesis of MC3T3-E1 showed significant association with the mammalian target of rapamycin (mTOR) phosphorylation by RA, which required further study about the mechanism. The approach to this modified Ti surface presented in this paper has high research value for the development of Ti-based implant.

Magnetic Hydroxyapatite Composite Nanoparticles for Augmented Differentiation of MC3T3-E1 Cells for Bone Tissue Engineering.

Project description:Progressive aging harms bone tissue structure and function and, thus, requires effective therapies focusing on permanent tissue regeneration rather than partial cure, beginning with regenerative medicine. Due to advances in tissue engineering, stimulating osteogenesis with biomimetic nanoparticles to create a regenerative niche has gained attention for its efficacy and cost-effectiveness. In particular, hydroxyapatite (HAP, Ca10(PO4)6(OH)2) has gained significant interest in orthopedic applications as a major inorganic mineral of native bone. Recently, magnetic nanoparticles (MNPs) have also been noted for their multifunctional potential for hyperthermia, MRI contrast agents, drug delivery, and mechanosensitive receptor manipulation to induce cell differentiation, etc. Thus, the present study synthesizes HAP-decorated MNPs (MHAP NPs) via the wet chemical co-precipitation method. Synthesized MHAP NPs were evaluated against the preosteoblast MC3T3-E1 cells towards concentration-dependent cytotoxicity, proliferation, morphology staining, ROS generation, and osteogenic differentiation. The result evidenced that MHAP NPs concentration up to 10 µg/mL was non-toxic even with the time-dependent proliferation studies. As nanoparticle concentration increased, FACS apoptosis assay and ROS data showed a significant rise in apoptosis and ROS generation. The MC3T3-E1 cells cocultured with 5 µg/mL MHAP NPs showed significant osteogenic differentiation potential. Thus, MHAP NPs synthesized with simple wet chemistry could be employed in bone regenerative therapy.

Effect of nano-hydroxyapatite and phosphate on thorium toxicity – Arabidopsis transcriptomic study

Project description:Thorium (Th) is a ubiquitous element but elevated concentrations caused by anthropogenic or natural activities pose health and environmental risks. Nowadays, Th is obtained mainly as by-product during the mining and extraction rare earth element. However, due to its abundance in nature, which is higher than the abundance of uranium, Th is considered as a potential fuel for the reactors. Therefore, in this study, we focused on the effect of thorium on Arabidopsis plants at the transcriptomic level. Since Th and phosphorus form insoluble phosphates, we focused on the effect of Th in the presence/absence of phosphate. Furthermore, we investigated the effect of hydroxyapatite nanoparticles, which are considered as material for in situ remediation, on Th toxicity. Not surprisingly, the highest number of DEGs was recorded in the combination of P absence and Th presence followed by the presence of Th in the standard hydroponic medium, the absence of P in the medium, the presence of nano-hydroxyapatite, and the presence of Th together with hydroxyapatite. Besides a high number of genes involved in stress response, Th also induced the transcription of genes involved in response to P starvation, indicating that Th decreased the availability of P. P deficiency caused by Th contributed to the phytotoxic effect of Th. Nano-hydroxyapatite significantly reduced Th toxicity. Nano-hydroxyapatite also served as a source of inorganic P for the plants, but caused deficiency of essential metals indicated by up-regulation of genes involved in their acquisition.

Integrating whole blood transcriptomic collection procedures into the current anti-doping testing system, including long-term storage and re-testing of anti-doping samples [Long-term]

Project description:Purpose: Recombinant human erythropoietin administration studies involving “omics” approaches have demonstrated a gene-expression signature that could aid detection of blood doping. However, current anti-doping testing does not involve blood collection into tubes with RNA preservative. This study investigated if whole blood in long-term storage could be used for transcriptomic analysis despite lacking RNA preservation. Methods: Whole blood samples were collected from thirteen male healthy individuals. Long-term storage: whole blood collected into Tempus™ tubes and K2EDTA tubes and subjected to long-term (i.e., −80°C) storage and RNA extracted. After storage, Tempus and K2EDTA tubes were thawed and extracted using Tempus™ Spin RNA Isolation Kit (Life Technologies, Carlsbad, CA, USA). Samples from seven subjects that presented higher RIN value (≥7) were selected for RNA_Seq analysis. Results: The experiment provided RNA quality and purity for gene expression analysis. Total of 19239 genes were mapped and the gene expression analysis showed that 658 genes were differentially expressed (which means 3.4% of mapped genes). With 269 being up-regulated and 389 down-regulated. None of the transcripts described in previous studies as biomarkers for blood doping (Durussel et al. 2016; Wang, Durussel, et al. 2017) were differently expressed. Conclusion: RNA quantity, purity and integrity was not significantly compromised from long-term storage in blood storage tubes lacking RNA stabilisation, indicating that transcriptomic/omics analysis could be conducted using anti-doping samples collected or biobanked without RNA preservation.

Cellular Mg2+ decrease causes a distinctive NF-κB-dependent form of cell death

Project description:Mg2+ is an essential cofactor for numerous enzymes, supporting fundamental cellular processes. Phosphatase of regenerating liver (PRL) protein family, inhibits cyclin M (CNNM) Mg2+ efflux transporters. To elucidate the physiological role of PRL in Mg2+ homeostasis at the cellular level, we employed combined genetic knockout and knockdown approaches. Such PRL KO + KD led to marked reduction of intracellular Mg2+ levels and triggered extensive cell death. To investigate the mechanism underlying cell death induced by PRL KO + KD, we next performed transcriptomic analysis to profile the changes in global gene expression, which revealed activation of the NF-κB pathway, and accordingly, genetic deletion of NF-κB p65 subunit abrogated cell death. Similarly, CNNM overexpression triggered intracellular Mg2+ decrease, NF-κB activation and subsequent cell death. Notably, this form of cell death exhibited unique morphological features, including actin-driven fiber-like protrusions, distinguishing it from known cell death modalities. Our findings uncover a previously unrecognized mode of NF-κB-dependent cell death triggered by intracellular Mg2+ decrease.
