Aplysia Pleural Ganglia 7 Days After Long-Term Sensitization Training.


ABSTRACT: We used a custom-designed microarray to characterize the transcriptional response that persists 5 days after long-term sensitization training in the marine mollusk Aplysia californica, a time-point at which recall has partially decayed but there is still clear sensitization memory expressed in most animals. Aplysia were exposed to a 1-day unilateral LTS training protocol (4 rounds of noxious shock at 30 min intervals, each shock 10s of 1/2s off 1/2on 90mA 60-hz biphasic square-wave shock). Duration of the tail-elicited siphon withdrawal reflex was measured before training, 1 day after training, and 5 days after training. From the 1-day post-tests we selected 32 animals showing strong sensitization responses (at least 30% increase in reflex duration on the side of training). At the 5-day post-test we observed substantial but not complete decay of the expresison of sensitizaiton. Plueral ganglia were harvested immediately after the 5-day post tests for microarray analysis; these ganglia contain the VC nociceptors known to help encode sensitization memory. Samples from a left-trained and right-trained animal were combined to smooth out lateralized gene expression (32 animals produced 16 microarray samples, each having a set of trained plueral ganglia and a set of untrained plueral ganglia). In addition, within each batch of animals collected we matched the highest-forgetting left-trained animal with the highest-forgetting right-trained animal, and so on, producing samples consistent in levels of forgetting and considerable diversity in forgetting across samples.
