Myeloid Piezo1 Drives Cardiac Repair Through Orai1–Rac1-Dependent Efferocytosis

ABSTRACT: Monocytes/macrophages orchestrate myocardial remodeling through efferocytosis post-myocardial infarction (MI). While the mechanosensitive channel Piezo1 is known to modulate calcium influx in cardiac and immune contexts, its myeloid-specific role in post-infarction healing remains elusive. Single cell RNA sequencing at 3 days post-MI revealed that the efferocytosis-associated genes were upregulated in Piezo1∆LysM group, which serve as a cardioprotective role.

Project description:CD40 expressed on the macrophage membrane is recognized as one of the characteristic biomarkers for their inflammatory phenotype. We demonstrated that macrophage CD40 functionally promotes post-myocardial infarction (MI) repair. To elucidate the role of CD40 in the post-MI repair process, we dissected infarct and border zones from the hearts of wild-type (WT) and CD40 knockout (KO) mice at 3 days post-MI. Mononuclear macrophages (CD45⁺CD11b⁺CD64⁺) were isolated via flow cytometry and subjected to single-cell RNA sequencing analysis. This study establishes that CD40 plays a critical role in macrophage efferocytosis. Furthermore, analysis of scRNA-seq data led to the identification of developmental precursor cells of repair-phenotype macrophages.

Project description:Study of the impact of 4-oxo-RA on LSK cell characteristics post-myocardial infarction in RarB KO mice. Myocardial infarction was induced in female RarB KO mice aged 6 to 12 weeks by LAD occlusion. On the 1st and 2nd post-MI days, mice were intraperitoneally injected with 30 mg/kg 4-oxo-RA (MI+4-oxo-RA) or DMSO in PBS (MI+vehicle). Three days post-MI, mice were euthanized. ScRNA-seq was performed on 25k LSK cells (Lin-, Sca1+, c-Kit+) that were facs sorted.

Gene expression profiling reveals potential prognostic biomarkers associated with the progression of heart failure

Project description:Heart failure (HF) is the most common cause of morbidity and mortality in the developed countries, especially considering the present demographic tendencies in those populations. We identified biologically relevant transcripts that are significantly altered in the early phase of myocardial infarction (MI) and are associated with the development of post-myocardial infarction HF. We collected peripheral blood samples from patients (n=111) with ST-segment elevation myocardial infarction (STEMI) at four time points (admission, discharge, 1 month after MI, and 6 months after MI). Control group comprised patients (n=46) with a stable coronary artery disease and without a history of myocardial infarction. Affymetrix HuGene 1.0 ST arrays were used to analyze mRNA levels in periperal blood mononuclear cells (PBMCs) isolated from the study and control groups. Samples from the first three time points were compared with the samples from the same patients collected 6 months after MI (stable phase) and with the control group. Additionaly, based on plasma NT-proBNP level and left ventricular ejection fraction parameters the STEMI patients were divided into HF and non-HF groups.We attempted to identify transcripts whose differential expression on the 1st day of myocardial infarction predicted which patients would develop symptoms of HF during the 6 months of follow-up. For this purpose, we compared the microarray results for samples collected on admission for the HF group versus the non-HF group.

Gene expression profiles at the single-nucleus level in heart tissue after myocardial infarction and saNppa-LNP treatment

Project description:This project aims to investigate the cellular and molecular mechanisms underlying the therapeutic effects of a self-amplifying RNA encoding atrial natriuretic peptide (saNppa) in myocardial infarction (MI). In this study, a myocardial infarction model was established in C57BL/6 mice by coronary artery ligation. On the day of surgery, mice received intramuscular injection of saNppa formulated in lipid nanoparticles (saNppa-LNP) or phosphate-buffered saline (PBS) as control. Single-nucleus RNA sequencing (snRNA-seq) was performed on left ventricular tissue collected at Day 7 post-MI and treatment. Three experimental groups were included: sham-operated mice, MI mice treated with PBS (MI+PBS), and MI mice treated with saNppa-LNP (MI+saNppa-LNP). This dataset provides a comprehensive single-nucleus transcriptional landscape of the post-infarct heart and enables cell type–specific analysis of transcriptional changes associated with myocardial infarction and saNppa self-amplifying RNA therapy.

Spatially-targeted optical microproteomics timecourse of rat myocardial infarction

Project description:Myocardial infarction (MI) results from occlusion of blood supply to the heart muscle causing death of cardiac muscle cells. Following myocardial infarction (MI), scar formation acts to mechanically stabilise the injured heart to prevent rupture and death. While fibroblasts and macrophages are implicated in post-MI scar formation and maturation, their exact contributions to this process are poorly characterised, especially in the long-term (i.e., beyond 1-week post-MI). Here, we employ state-of-the-art spatially-targetted optical micro-proteomics (STOMP) to isolate proteomes of tissue fractions enriched for fibroblasts (sma+) and macrophages (cd68+) over a 6-week post-MI timecourse and compare them to whole-scar proteome. We illustrate dynamic, specific changes in sma- and cd68-enriched fraction protein composition over 1-6 weeks post-MI with some of these changes reflected in whole-infarct preparations. These results link specific cell populations to specific protein changes in whole infarct.

Effect of Card9 knockout on gene expression in cardiac tissues of mice post myocardial infarction

Project description:Immune cell infiltration in response to myocyte death contributes to extracellular matrix (ECM) remodeling and scar formation after myocardial infarction (MI). Caspase-recruitment domain protein 9 (CARD9) which belongs to CARD family acts as an adapter that mediate the transduction of proinflammatory signaling cascades in innate immunity. To investigate the role of CARD9 in cardiac injury and repair post ischemia, we subjected Card9 knockout mice to myocardial infarction (MI) , and then performed RNA-seq and gene expression profiling analysis using the ischemic cardiac tissues at 3 days post-MI, to identify key genes and pathways regulated by CARD9.
