Mitochondrial Iron Sensitizes Senescent Cancer Cells to Ferroptosis

ABSTRACT: The chemotherapy regimen FOLFIRINOX has been shown to induce senescence in pancreatic ductal adenocarcinoma (PDAC) cells, as well as in immortalized normal human pancreatic ductal epithelium (HPNE-hTERT) cells. To investigate the transcriptional changes associated with treatment-induced senescence in non-malignant cells, RNA sequencing was performed on HPNE-hTERT cells that were either untreated (vehicle control) or treated with FOLFIRINOX for 4 days.

Project description:FOLFIRINOX induces senescence in pancreatic tumor cells. However, some cells can spontaneously escape this senescence. We wanted to characterize the escape mechanisms from FOLFIRINOX-induced senescence of pancreatic tumor cells. Our in vitro adaptation model to FOLFIRINOX shows that cells that have already escaped 5 treatment cycles become less sensitive to FOLFIRINOX.

Senescent cancer-associated fibroblasts in pancreatic adenocarcinoma impair CD8+ T cell activation and limit responsiveness to immunotherapy in mice

Project description:Senescent cells within tumors and their stroma exert complex pro- and anti-tumorigenic functions, yet the potential for targeting such senescent cells for improved therapy remains largely unknown. Here, we uncover the presence of a senescent subset of cancer-associated fibroblasts (CAFs) within pancreatic ductal adenocarcinomas (PDAC) and in premalignant lesions in mice and humans. Senescent CAFs represented different previously-defined CAF subtypes. Senescent CAFs isolated from mouse and humans expressed elevated levels of immunoregulatory genes. Depletion of senescent CAFs, either genetically or using the Bcl-2 inhibitor ABT-199 (venetoclax), increased the proportion of activated CD8+ T cells in mouse pancreatic carcinomas, whereas induction of CAF senescence had the opposite effect. Combining ABT-199 with an immune checkpoint therapy regimen significantly reduced mouse tumor burden. These results indicate that senescent CAFs in PDAC stroma limit the numbers of activated cytotoxic CD8+ T cells, and suggest that their targeted elimination through senolytic treatment may enhance immunotherapy.

Investigation of the rhythmic changes in the proteome of different subtypes of pancreatic ductal adenocarcinoma

Project description:At present, there is no reliable biomarker for the diagnosis of pancreatic ductal adenocarcinoma (PDAC). Due to this, together with the asymptomatic nature of the cancer, PDAC is often diagnosed at an advanced stage with an abysmal 5-year survival rate. Hence, a reliable biomarker for the timely diagnosis of PDAC is essential. The altered levels of protein expression are one of the hallmarks of cancer. Understanding these changes in PDAC could yield insights into the development of the disease and hence could potentially identify new biomarkers. Hence, in this study, mass spectrometry-based label‐free quantitative proteomics was employed to investigate the differently expressed proteins in the pancreatic cancer cell lines (Capan-2 and PANC-1), benign pancreatic cells (hTERT-HPNE) and normal pancreatic duct epithelial cells (H6c7). Furthermore, to study the circadian variations in the secretome of the pancreatic cell lines, the rhythmic expression of proteins was investigated at 6-hour intervals over 24 hours. The findings of this study provide novel insights into the processes and pathways involved in PDAC, potentially leading to the identification of circadian-based subtype-specific biomarkers.

miRNA Expression in Senescent and Immortalized BJ fibroblasts

Project description:Limited data exists regarding changes of microRNA (miRNA) expression during senescence in human cells and no reports correlate telomerase expression with regulation of senescence-related miRNAs. We used miRNA microarrays to provide a detailed account of miRNA profiles for early passage and senescent human foreskin (BJ) fibroblasts as well as early and late passage immortalized fibroblasts (BJ-hTERT) that stably express the human telomerase reverse transcriptase subunit hTERT. The revelation that miRNA expression changes with extended passaging in BJ-hTERT cells will contribute to a comprehensive understanding of the connections between telomerase expression, senescence and processes of cellular aging.

Patient-derived organoids after neoadjuvant FOLFIRINOX predict therapy resistance in pancreatic ductal adenocarcinoma

Project description:Pancreatic ductal adenocarcinoma is an extreme lethal cancer with a poor treatment response to all the current chemotherapies. We generated a library of PDAC organoid lines from resected tumors, five from treatment-naive patients and five from patients who received neoadjuvant FOLFIRINOX (eight cycles). We assessed weather residual cancer cells in the tumor lesions of the patients treated with neoadjuvant FOLFIRINOX developed resistance to the treatment. Further we perfromed transcriptome analysis to investigate: 1- weather organoids recapitualting their corresponding tumor tissue 2- To investigate any modifications in the expression of the genes inflicted by FOLFIRINOX treatment.

Prox1 haploinsufficiency sensitizes the pancreas to KrasG12D-induced transformation

Project description:Oncogenic mutations in Kras initiate neoplastic transformation in the pancreas through a process that hijacks the activity of developmental regulators and induces an inflammatory microenvironment. We report that the homeodomain transcription factor Prox1 is a novel component of a progenitor signature that is activated in acinar cells undergoing dedifferentiation and ductal metaplasia conversion. Also, the conditional deletion of a single Prox1 allele markedly accelerates early transformation and significantly enhances features of inflammation in pancreatic tissues carrying a Kras oncogene. By using in vitro and in silico approaches, we demonstrate that Prox1 negatively regulates the expression of proinflammatory genes and genes implicated in malignant transformation in pancreatic cells. Microrrays were used to identify gene expression profiles and pathways that are differentially activated after enforced expression of PROX1 in the PROX1-negative human pancreatic ductal adenocarcinoma (PDAC) cell line Capan1. MSCV retroviral transduction was used to generate Capan1 cells that ectopically express PROX1 and selected for puromycin resistance. Three independent transductions with control virus (MSCV_puro) and 3 independent transductions with Prox1-cDNA virus (MSCV_PROX1_puro) were conducted for these experiments.

Targeting ribosome biogenesis reinforces ERK-dependent senescence in pancreatic cancer

Project description:Aberrant activation of the ERK signaling pathway triggers a protective anticancer response characterized by stable growth arrest and activation of tumor suppressors called cellular senescence. Pancreatic adenocarcinomas (PDAC) often possess mutations in K-Ras that activate the ERK pathway. Pancreatic intraepithelial neoplasia of low degree display high levels of phospho-ERK consistent with senescence acting as a barrier for malignant transformation. However, advanced lesions downregulate phospho-ERK levels circumventing the senescence barrier. Restoring ERK hyperactivation in PDAC using an activated allele of the kinase RAF, leads to ERK-dependent growth arrest with senescence biomarkers. Phosphoproteomics analysis of ERK-dependent senescence in PDAC revealed a decrease in several nucleolar phosphoproteins suggesting that high levels of ERK lead to senescence via nucleolar stress. Consistent with this explanation, ERK-dependent senescent cells displayed intranucleolar foci containing RNA polymerase I. Combining ribosome biogenesis inhibitors with ERK hyperactivation reinforced the senescence response of PDAC cells. The drug cocktail FOLFIRINOX, currently the best treatment for PDAC, also triggered ERK hyperactivation and nucleolar stress characterized by nucleolar foci, solid amyloid aggregates and a decrease in 5.8S and 28S rRNAs. We thus suggest that drugs targeting ribosome biogenesis can improve the senescence anticancer response in pancreatic cancer.
