RNA sequencing of polysome-associated transcripts in EIF4A1-overexpression and mock-virus APOE4 NGN2 neurons

ABSTRACT: Polysome-associated RNA was profiled in human NGN2-induced neurons of APOE4 genotype after mock-virus treatment or EIF4A1 overexpression. Neurons were co-plated with rat astrocytes, harvested at 7 weeks post-differentiation, and RNA was isolated from pooled heavy polysome fractions (fractions 17, 18, and 19) following sucrose-gradient fractionation. Libraries were prepared after poly(A) enrichment using SMART-Seq v4 with tagmentation and sequenced as 150-bp paired-end reads on an Illumina NovaSeq X Plus platform.
