RNA-seq of Smc3-GFP flox/flox mouse embryonic stem cells with conditional knockout

ABSTRACT: To investigate the functional consequences in the absence of Smc3 in mouse embryonic stem cells (mESCs), we treated the Smc3 GFP flox/flox mESCs with 4-OHT to deplete the Smc3 alleles, and then performed RNA-seq on two independent Smc3 flox/flox GFP ESC lines before and after Cre-mediated excision. RNA-seq analysis revealed dysregulation of key genes governing pluripotency and lineage commitment, highlighting the indispensable role of cohesin in preserving the transcriptional identity of embryonic stem cells.
