Single-Cell RNA Sequencing Reveals Yiqi Huayu Jiedu Formula Alleviates Acute Respiratory Distress Syndrome Through Regulation of Macrophage Chemotaxis

ABSTRACT: Integrated network pharmacology, single-cell RNA sequencing, and experimental analyses revealed that Yiqi Huayu Jiedu Formula (YQHYJDF) protects lung tissue by inhibiting macrophage chemotaxis and neutrophil recruitment through suppression of proinflammatory cytokines IL-6, IL-1β, and TNF-α, PI3K/AKT, MAPK, and NF-κB signaling, and downregulation of chemokines CCL2, CCL7, and CCL12.

Project description:Connections between inflammation and gene-network regulation are suggested important in understanding the therapeutic target of stroke and in illuminating underlying mechanism. However, studies on the establishment of network relating with inflammation during stroke process are still in their early stages. We selected the inflammation genes from the whole mRNA expression results. And after a series of analysis, we found the promising target genes of Yiqi Jiedu formulae used in clinic for stroke, which might give an explanation to the common characteristic about TCM treatment called PK-PD inconsistent.

Project description:This study employed RNA-seq to profile the transcriptomic changes in the renal cortex of diabetic nephropathy (DN) mouse models and to investigate the regulatory effects of the traditional Chinese medicine formula Yiqi Huoxue Yangyin Decoction (YHY). Kidney cortex tissues were collected from diabetic db/db mice and non-diabetic db/m control mice. Db/db mice were further divided into two groups: one treated with YHY and one vehicle-treated. Project description:In M-CSF–derived macrophages, bulk RNA-seq was performed on five biological replicates per cytokine condition (IL‑4, IFN‑γ, IL‑10, TGF‑β). IL‑4 dominated PC1 (54% variance) driving a reparative module with EMT, UPR, and mTORC1 signatures, while IFN‑stimulated genes (Gbp4, Ifi44) were repressed. PC2 (26% variance) spanned an inflammatory‑to‑remodeling continuum: IFN‑γ anchored the chemokine‑rich, IFN‑responsive pole (Ccl7, Ccl2, Ccl12), whereas TGF‑β anchored a matrix‑remodeling/angiogenic pole (Mmp14, Cav1, Cspg4); IL‑10 shifted modestly toward the IFN‑low quadrant.
