Single-cell transcriptomic landscape of ketone body–mediated modulation of renal mTORC1 signaling in proximal tubule–specific Tsc1 knockout mice

ABSTRACT: We performed single-cell RNA sequencing of mouse kidneys to investigate the effects of ketone bodies on renal mTORC1 signaling. A proximal tubule–specific Tsc1 knockout model (PTC- iKO) was used to induce constitutive mTORC1 activation. 1,3-butanediol, a precursor of ketone bodies that elevates circulating ketone levels, was administered as a dietary supplement. Three groups were analyzed: Tsc1 flox controls, Tsc1 PTC-iKO under a normal diet, and Tsc1 PTC-iKO with 1,3-butanediol treatment. This dataset enables cell type–specific analysis of transcriptional changes associated with mTORC1 activation and ketone body exposure in the kidney.
