Multitissue Single-Cell Analysis Reveals Differential Tissue, Cellular, and Molecular Sensitivity between Fructose and High fat high sucrose Diets

Project description:Metabolic syndrome (MetS) is a complex disorder with multidimensional etiology that encompasses diverse symptoms such as hyperlipidemia, abdominal obesity, and insulin resistance. Western diets such as the high fat high sucrose diet (HFHS) and those high in fructose ave been associated with increased prevalence of MetS. Despite the fact that various metabolic tissues have been implicated in MetS pathogenesis, the role of individual cell types embedded in these tissues has yet to be elucidated. To address this, we performed single cell RNA sequencing to examine thousands of individual cells from the hypothalamus, liver, adipose, and small intestine from both HFHS- and fructose-induced MetS mouse models. We found differential sensitivity of responsive cell types, genes, and pathways between HFHS and fructose diets, with hypothalamic neurons particularly sensitive to the high fructose diet and adipose progenitor cells particularly sensitive to HFHS diet. Network analysis identified both known (Avp, Apoe, C3) and novel ligands (Gal and Fga) that mediate ligand-receptor crosstalk between tissues in MetS. The identification of major cell types, molecular pathways, and regulators of MetS induced by different risk diets facilitates precision treatment of MetS subtypes.

Gene expression data from FLCN-restored UOK257-2 renal cancer cells

Project description:UOK257 cell line was derived from a BHD patient. It harbors a germline mutation in FLCN (c.1285dupC) and LOH. UOK257-2 cells were generated from UOK257 cells by introducing wildtype FLCN using retrovirus. FLCN inactivation induces TFE3 transcriptional activity by increasing its nuclear localization. Thus expression microarray was used to identify the genes regulated by FLCN and TFE3. UOK257-2 cells expressing wildtype FLCN were transfected with 1) scrambled, 2) FLCN, 3) TFE3 and 4)FLCN/TFE3 siRNAs with Lipofectamine 2000 transfection reagent. Cells were harvested 3 days after transfection and RNAs were isolated using Trizol reagent and RNeasy Mini Kit. The genes induced by FLCN siRNA and reduced by TFE3 siRNA were chosen as a candidate genes regulated by FLCN and TFE3.

High Fructose-Driven Epigenetic and Metabolic Reprogramming in Glomerular Podocyte

However, the underlying pathological mechanisms of fructose-induced podocyte injury remain elusive. In this study, we have used an iTRAQ based quantitative proteomic strategy to comprehensively characterize the dynamic proteome changes in glomeruli of high fructose-fed rats, and revealed global fructose-induced glomerular metabolic reprogramming at four different stages during the progression of high fructose modeling.

Gene expression data from liver of rat fed a high-fat diet and high-fructose water without/with Eucommia ulmoides leaves

Project description:To investigate effects of intake of Eucommia ulmoides leaves on hyperlipidemia, we performed gene expression profiling on rat liver by microarray analysis. Microarray analysis revealed that Eucommia ulmoides leaves up-regulated the gene expression involved in alpha-, beta-, and omega-oxidation of fatty acids, mainly relating to peroxisome proliferator-activated receptor signaling pathway. Rats were fed a high-fat diet and high-fructose water without/with orally administration of Eucommia ulmoides leaves for 5 weeks. Livers were taken for RNA extraction and hybridization on Agilent microarrays.

Gene expression altered by high-fructose diet feeding and Chrebp deficiency

Project description:Epidemiologic and animal studies implicate overconsumption of fructose in the development of non-alcoholic fatty liver disease, but the molecular mechanisms underlying fructose-induced chronic liver diseases remains largely unknown. We present evidence supporting the essential function of the lipogenic transcription factor ChREBP in mediating adaptation response to fructose and protecting against fructose-induced hepatotoxicity. High-fructose diet (HFrD) activates hepatic lipogenesis via a ChREBP-dependent manner in wildtype mice, while inducing steatohepatitis in Chrebp-KO mice. In Chrebp-KO mouse livers, HFrD reduces levels of molecular chaperones and activates the CHOP-dependent unfolded protein response, whereas administration of chemical chaperone or Chop shRNA rescues liver injury. Gene expression profiling revealed elevated expression of cholesterol biosynthesis genes in Chrebp-KO livers after HFrD, in parallel with increased abundance of nuclear SREBP2. genes expression were compared between livers of wildtype mice fed 70%-fructose-diet v.s. regular chow, and between livers of Chrebp-/- mice v.s. wildtype mice fed 70%-fructose-diet.

High-Fructose Corn Syrup Enhances Intestinal Tumor Growth in Mice

Project description:Excessive consumption of beverages sweetened with high-fructose corn syrup (HFCS) is associated with obesity and with an increased risk of colorectal cancer. Whether HFCS contributes directly to tumorigenesis is unclear. We investigated the effects of daily oral administration of HFCS in APC mutant mice, which are predisposed to develop intestinal tumors. The HFCS-treated mice showed a dramatic increase in tumor size and tumor grade in the absence of obesity or metabolic syndrome. HFCS increased the levels of fructose and glucose in the intestinal lumen and serum, respectively, and the tumors absorbed both sugars. Within the tumors, fructose was converted to fructose-1-phosphate, leading to activation of glycolysis and increased synthesis of fatty acids that support tumor growth. These mouse studies support the hypothesis that the combination of dietary glucose and fructose, even at a moderate dose, can enhance tumorigenesis.

Analysis of differentially expressed genes revealed sirtuin signaling and a network of genes regulated by the peroxisome proliferator activated receptor alpha were significantly altered in response to the high fructose diet.
