Leveraging TNFR2 for antitumour immunity: Treg depletion and myeloid reprogramming versus T cell costimulation

ABSTRACT: We conducted single-cell RNA sequencing using the 10x Genomics Chromium Next-GEM Flex protocol to investigate the mechanism of action of two TNFR2-targeting antibodies—one ligand-blocking and the other agonistic. Our goal was to assess their anti-cancer potential in comparison to established immune checkpoint inhibitors, anti-PD1 and anti-CTLA4. These studies provide new insights and role of TNFR2 as a therapeutic target in cancer.
