Plasma extracellular vesicle miRNA profiling in gastric cancer patients and healthy donors

ABSTRACT: Extracellular vesicles were isolated from plasma samples of gastric cancer patients and healthy donors. EV-associated miRNAs were profiled by small RNA sequencing to identify gastric cancer-associated EV-miRNA signatures.

Project description:Extracellular vesicles are structures surrounded by a lipid bilayer that facilitate intercellular communication by transporting biomolecules. These vesicles are now commonly referred to as part of liquid biopsy. In this study, we examine the characterization of miRNAs found in extracellular vesicles from patients with both benign gastric diseases and gastric cancer. By studying these miRNAs, we aim to identify potential biomarkers for gastric cancer.

Project description:Metastasis is a major problem of gastric cancer. In this study, small extracellular vesicle (sEV)-derived miRNAs were sequenced to screen biomarkers for GC’s organo-tropic metastasis. Plasma from 40 treatment-naïve gastric cancer patients including 10 no metastasis (M0) and 30 distant metastasis (M1) were assessed by sEV-miRNA-sequencing. sEV miRNAs with diverse expression profiles across different metastatic patterns were combined into sigantures to characterize and predict gastric cancer metastasis.

Plasma extracellular vesicle miRNA profiling in cervical cancer patients and healthy controls

Project description:Cervical cancer (CC) is a malignancy characterized by persistent HPV infection and tumor microenvironment remodeling. To explore the molecular characteristics of plasma extracellular vesicles (EVs) in CC, we performed miRNA sequencing on plasma EVs from 3 CC patients and 3 healthy controls. Differentially expressed miRNAs were identified, revealing potential biomarkers and providing insights into early diagnosis and therapeutic intervention.

Unlocking the diagnostic power of plasma extracellular vesicle miR-200 family in pancreatic ductal adenocarcinoma

Project description:Background Distinguishing benign from malignant pancreaticobiliary disease is challenging because of the absence of reliable biomarkers. Circulating extracellular vesicles (EVs) have emerged as functional mediators between cells. Their cargos, including microRNAs (miRNAs), are increasingly acknowledged as an important source of potential biomarkers. This multicentric, prospective study aimed to establish a diagnostic plasma EV-derived miRNA signature to discriminate pancreatic ductal adenocarcinoma (PDAC) from benign pancreaticobiliary disease. Methods Plasma EVs were isolated using size exclusion chromatography (SEC) and characterised using nanoparticle tracking analysis, electron microscopy and Western blotting. EV-RNAs underwent small RNA sequencing to discover differentially expressed markers for PDAC (n = 10 benign vs. 10 PDAC). Candidate EV-miRNAs were then validated in a cohort of 61 patients (n = 31 benign vs. 30 PDAC) by RT-qPCR. Logistic regression and optimal thresholds (Youden Index) were used to develop an EV-miR-200 family model to detect cancer. This model was tested in an independent cohort of 95 patients (n = 30 benign, 33 PDAC, and 32 cholangiocarcinoma). Results Small RNA sequencing and RT-qPCR showed that EV-miR-200 family members were significantly overexpressed in PDAC vs. benign disease. Combined expression of the EV-miR-200 family showed an AUC of 0.823. In an independent validation cohort, application of this model showed a sensitivity, specificity and AUC of 100%, 88%, and 0.97, respectively, for diagnosing PDAC.

EVs were separated from plasma of sepsis patients at admission as well as healthy controls. The expression of EV-miRNAs was evaluated by microarray and qRT-PCR.

PSAT1 drives extracellular vesicle release in multiple types of cancer and promotes bone metastasis by activating osteoclasts

Project description:Cancer cells secrete extracellular vesicles (EVs) to regulate cells in the tumor microenvironment for their own benefit to grow and survive in the patient’s body. Although emerging evidence has demonstrated the molecular mechanisms of EV release, regulating cancer-specific EV secretion remains challenging . In this study, we applied a microRNA (miRNA) library to reveal the universal mechanisms of EV secretion in cancer cells. Here, we identified miR-891b and its direct target gene, phospho-aminotransferase 1 (PSAT1), which promotes EV secretion through the serine-ceramide synthesis pathway. Inhibition of PSAT1 affected EV secretion in multiple types of cancer, suggesting that the miR-891b/PSAT1 axis shares a common mechanism of cancer EV secretion.

Non-canonical small noncoding RNAs in the Plasma extracellular vesicles as novel biomarkers in gastric cancer

Project description:Identifying robust diagnostic biomarkers for gastric cancer (GC) remains a significant challenge. Emerging studies highlight extracellular vesicle (EV)-derived RNAs in cancer biology, but the diagnostic potential of circulating EV-derived small non-coding RNAs (sncRNAs) in GC is poorly understood. Using panoramic RNA display by overcoming RNA modification aborted sequencing (PANDORA-seq), we mapped non-canonical sncRNAs—specifically ribosomal RNA-derived small RNAs (rsRNAs) and transfer RNA-derived small RNAs (tsRNAs)—in plasma EVs. We identified a three-rs/tsRNA signature that discriminates GC patients from healthy individuals with high sensitivity (80.42%) and specificity (87.43%) (143 GC vs 167 controls). For early_x0002_stage GC (stage I), sensitivity and specificity were 81.97% and 81.44%, respectively. Furthermore, the three_x0002_rs/tsRNA signature was evaluated in two independent cohorts, resulting in AUC values of 0.97 and 0.91 for distinguishing GC from healthy controls. Functional analyses revealed that these rs/tsRNAs regulate the ErbB/Hippo pathways, suggesting them in the underlying pathogenesis andtherapeutic potential. This study establishes a novel EV-derived sncRNA signature for early GC detection.
