Genome-wide H3K27ac profiling of RAW264.7 macrophages treated with LPS and CCS1477

ABSTRACT: Histone H3 lysine 27 acetylation (H3K27ac) ChIP-seq was performed to investigate genome-wide epigenetic changes in RAW264.7 macrophages following inflammatory stimulation and EP300 inhibition. RAW264.7 macrophages were pretreated with the EP300 inhibitor CCS1477 for 2 hours and subsequently stimulated with lipopolysaccharide (LPS) in the presence or absence of CCS1477 for 6 hours before chromatin immunoprecipitation and sequencing. Genome-wide H3K27ac occupancy was analyzed to characterize changes in chromatin activation associated with inflammatory responses and EP300 inhibition.
