Time-dependent BMP4 signaling directs lineage specification in human mesoderm

ABSTRACT: We investigated the mechanisms underlying mesodermal heterogeneity using a minimal directed differentiation system that simultaneously generates paraxial, intermediate and lateral plate mesoderm populations. Single-cell RNA sequencing across defined time points during hPSC differentiation (from day (D)3 to D9) revealed a temporal sequence of lineage specification with the lateral plate mesoderm emerging first, followed by paraxial and intermediate mesoderm differentiation.
