Genomic profiling of estrogen-related receptor identifies adult adipocyte-specific targets in Drosophila

ABSTRACT: Estrogen-related receptor (ERR) is a conserved nuclear receptor required for energy homeostasis. Here, we took a modified DamID genomic profiling approach (NanoDam) to identify ERR-specific targets in adult Drosophila male and female adipocytes. We found that ERR binds to the loci of genes encoding enzymes involved in glycolysis, the pentose phosphate pathway, and beta-oxidation.

The Drosophila Estrogen-Related Receptor acts within the larval fat body to promote triglyceride storage

Project description:Members of the Estrogen-Related Receptor (ERR) family of nuclear receptors (NR) serve a key role in coordinating triglyceride (TAG) accumulation with juvenile growth and development. In both insects and mammals, loss of ERR activity disrupts (TAG) storage during the post-embryonic growth phase, with loss-of-function mutations in mouseErraandDrosophila melanogaster dERRinducing a lean phenotype. However, the role of insect ERRs in controlling TAG accumulation remains poorly understood, as previous multiomic studies largely relied on whole animal analyses. Here we address this shortcoming by using tissue-specific approaches to determine how dERR regulates lipid metabolism within theDrosophilalarval fat body. We found that dERR autonomously promotes TAG accumulation within fat body cells by regulating expression of genes involved in glycolysis, the pentose phosphate pathway, lipid synthesis, andb-oxidation. As an extension of these results, we not only discovered thatdERRmutant fat bodies exhibit decreased expression of known HNF4 target genes but also found that dHNF4 activity is decreased indERRmutants. Overall, our findings indicate that dERR plays a multifaceted role in the larval fat body to coordinate lipid storage with developmental growth and hint at a conserved mechanism by which ERR and HNF4 homologs coordinately regulate metabolic gene expression

Project description:Estrogen-related receptor (ERR) beta is critical for maintenance of mouse embryonic stem cell (ESC) self-renewal under 2-inhibitor (“2i”) conditions. On the other hand, we biochemically revealed ERR activated transcription through the interaction with DNA, TFIIA, and C-terminal AF2 domain binding coactivators. To evaluate the biological significance of this finding, we analyzed the transcriptome of DNA-, TFIIH-, and coactivator-binding deficient mutant ERR beta-expressing ERR beta-knockout ESCs which showed compromised self-renewal activity. The results demonstrated that each interaction was required for regulating the specific biological/functional gene set among ERR beta-target genes.

NanoDam Profiling of EGL-43 in the Anchor Cell

Project description:Depending on the cellular context, TF expression can vary dramatically both spatially and temporally. These differences in expression patterns can result in tissue-specific differences in TF binding to downstream targets. To identify targets on a tissue-specific basis, Targeted DamID (TaDa) has been recently introduced to generate TF binding profiles in various models including C. elegans. However, TaDa suffers from portability such that a new promoter-TF fusion transgene must be constructed for every new experimental condition of interest. Here, we adapt NanoDam for usage in C. elegans, which relies on the use of endogenous TF-GFP knock-ins, a plethora of which have already been generated by the community. We report that NanoDam single copy transgenes consisting of lowly expressed, tissue-specific GFP nanobody-Dam fusions, when combined with endogenous GFP-tagged alleles of TFs, results in robust, tissue-specific profiling. Using an endogenous GFP-tagged allele of EGL-43/EVI1, we performed NanoDam profiling of two disparate tissue types, the anchor cell (AC) and dopaminergic neurons, and identify targets unique to each and shared by both cell types. We also identify two GATA TFs, ELT-6 and EGL-18, as novel regulators of AC invasion. Taken together, we demonstrate that NanoDam is capable of profiling endogenous GFP-tagged TFs to identify novel downstream targets in specific cell types of C. elegans.

Finding binding sites for nuclear receptors in S2 Schneider cells

The whole-genome experiments revealed that the sites of EcR receptor are partially overlapped with ERR binding sites. As ERR receptor specifically binds regulatory regions of glycolytic genes and genes of glycogen metabolism, the presence of EcR on ERR targets signifies involvement of the ecdysone signaling in regulation of carbohydrate metabolism.

Project description:DY131 is a pharmacological agonist of the orphan receptor estrogen-related receptor (ERR) γ which plays a crucial role in regulating energy generation, oxidative metabolism, cell apoptosis, inflammatory responses, etc. However, its role in acute liver injury is unknown. In the study, we evaluated the effect of DY131 on lipopolysaccharide (LPS)-induced liver injury. Transcriptomics analysis revealed that the dysregulated pathways associated with inflammation and metabolism were significantly reversed by DY131 in LPS-treated mice, providing more evidence in favor of the protective effect of DY131 against LPS-induced liver injury.

Differential gene expression between ERRa KO and WT mouse kidneys

Project description:Estrogen-related receptor (ERR) alpha is an orphan nuclear receptor highly expressed in the kidneys. ERRalpha is implicated in renal sodium and potassium homeostasis and blood pressure regulation. We used microarray analysis to identify differentially expressed genes in ERR alpha knockout mice kidneys versus wild-type. The results provide insight on the roles of ERRalpha in the kidney. Three biological replicates of WT and ERRaKO were performed, for a total of 6 samples. 2-3 month old males of each genotype were used.
