Chronic hypobaric hypoxia induces exposure-duration-dependent transcriptomic remodeling in the mouse cochlea

ABSTRACT: To investigate cochlear transcriptomic responses to chronic hypobaric hypoxia, male C57BL/6J mice were assigned to an ambient-pressure control group or exposed to simulated high-altitude hypobaric hypoxia for 2, 4, or 6 weeks. A staggered-entry design was used so that all groups underwent terminal cochlear tissue collection on the same day. Bulk RNA sequencing was performed on 20 independent cochlear samples, with five biological replicates per group. The study identified exposure-duration-associated transcriptional changes involving mitochondrial and intermediary metabolism, lipid metabolism, vascular/barrier-related signatures, and immune-related transcriptional features.
