Single-Cell Transcriptomic Profiling of Immune Cells from Parental and EMP1-Deficient MB49 Bladder Tumors.

ABSTRACT: In this study, we employed a single-cell RNA sequencing (scRNA-seq) approach using the 10x Genomics platform to investigate the immune microenvironment of syngeneic mouse bladder tumors. Tumors were established in immunocompetent C57BL/6 mice using parental or EMP1-knockout MB49 bladder cancer cells. Tumor-infiltrating immune cells were isolated and profiled at single-cell resolution to define immune cell populations, transcriptional states, and functional programs within the tumor microenvironment. Comparative analyses were performed to determine how tumor-cell EMP1 expression influences immune cell composition, activation and exhaustion states, inflammatory signaling, and intercellular communication. This dataset provides a single-cell transcriptomic resource for investigating EMP1-mediated regulation of antitumor immunity in bladder cancer.
