Transcriptome profiling of the spinal dorsal horn in a rat bone cancer pain model (Sham vs. BCP, part of a study on dexmedetomidine and the Nrf2/Slc7a11/ferroptosis pathway)

ABSTRACT: Bone cancer pain (BCP) is a major source of cancer-related suffering and is difficult to manage with opioid-based therapy. This dataset contains bulk RNA-seq data of the spinal dorsal horn from a rat BCP model, generated as part of a study investigating whether dexmedetomidine alleviates BCP by inhibiting spinal ferroptosis via the Nrf2/SLC7A11 pathway. BCP was induced by injecting Walker 256 mammary gland carcinoma cells into the left tibial bone marrow cavity of female Sprague-Dawley rats; sham-operated rats receiving inactivated Walker 256 cells served as controls (n = 3 per group). Spinal dorsal horn tissues were collected 15 days after inoculation. Compared with the sham group, 1072 genes were differentially expressed in the BCP group (848 up-regulated and 224 down-regulated; |log2 fold change| > 0.5, P < 0.05), with Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis showing significant enrichment of the ferroptosis pathway. These data identify ferroptosis as a core pathogenic pathway in the spinal dorsal horn of BCP rats.
