Single-cell RNA sequencing of human endothelial cells and macrophages in control and senescent endothelial cell co-cultures
Ontology highlight
ABSTRACT: Single-cell RNA sequencing was performed to characterize the transcriptional profiles and cellular heterogeneity of human endothelial cells and monocyte-derived macrophages following 3 days of three-dimensional co-culture. Control (young) and replicatively senescent endothelial cells were co-cultured with human macrophages in endothelial cell–macrophage spheroids. Single-cell transcriptomic profiling was used to identify major endothelial and macrophage populations and to characterize cell-state-specific gene expression patterns. Cell–cell communication analysis was subsequently performed using CellChat to investigate potential intercellular signaling networks within the co-culture system. The research has been supported by a grant No. 2021/43/B/NZ5/03336 from the National Science Center, Poland.
