Project description:We analyzed global mRNA expression in urogenital ridges of embryos that were either heterozygous for the MM-CM-<llerian Inhibiting Substance type II receptor or homozygously deleted to study the genes regulated by MIS during MM-CM-<llerian duct regression. Approximately 200 mosue embryos were sorted for heterozygous or homozygous deletion of the MISR2, urogenital ridges were collected and the gonads were discarded. mRNA was prepared from pooled ridges and sent for Affymetrix analysis.

Project description:The goal of this study was to identify potential AMH-induced genes and regulatory networks controlling regression by RNA-Seq transcriptome analysis of differences in Müllerian Duct mesenchyme between males (AMH signaling on) and females (AMH signaling off) in purified fetal Müllerian Duct mesenchymal cells. This analysis found 82 genes up-regulated in males during MD regression and identified Osterix (Osx)/Sp7, a key transcriptional regulator of osteoblast differentiation and bone formation, as a novel downstream effector of AMH signaling during MD regression.

Project description:The Wilms tumor protein Wt1 is a transcription factor known to play an important role in urogenital development. Mutations in the human Wt1 encoding gene (WT1) lead to several syndromes associated with defective renal and sexual development, namely WAGR-, Denys-Drash-, and Frasier-syndrome. During mammalian embryogenesis, urogenital development starts with formation of the urogenital ridges, consisting of the embryonic kidneys, called mesonephroi, and the gonads. In Wt1 knockout mice, these ridges start to form, degenerate, however, during further embryonic development . Although WT1 and the syndromes associated with its mutation are known for several years, it is still unclear which genes, regulated by Wt1, mediate the whole variety of Wt1's function in gonad and mesonephros development. To obtain a comprehensive view on Wt1-dependent gene activity in developing urogenital ridges, we employed microarray analysis. Comparison of gene expression in Wt1 wild-type and knockout mice, led to the identification of almost 150 genes with differences in expression levels higher than factor three. Project description:In an XX embryo, the Wolffian duct regresses under the control of the mesenchymal transcription factor COUP-TFII. To understand cellular and molecular actions underlying Wolffian duct regression, we preformed transcriptomic analyses of XX mesonephroi with or without Coup-tfII and genome-wide analysis of COUP-TFII chromatin occupancy in XX mesonephroi. The integrative analysis of COUP-TFII genome-wide binding and transcriptomic analysis revealed the suppression of muscle differentiation and extracellular matrix genes by COUP-TFII and identified a group of COUP-TFII’s potential transcriptional partners in the mesenchyme that potentially facilitate Wolffian duct regression. These findings provide insights into the molecular action of COUP-TFII in the Wolffian duct mesenchyme and identify a list of biologically relevant candidate genes and pathways for future functional analyses in sexual differentiation of reproductive tracts.

Project description:Precursors for the male and female reproductive tracts (also known as the Wolffian duct and the Müllerian duct) co-exist in a mammalian embryo before sexual differentiation. The Wolffian duct and Müllerian duct are surrounded by their own mesenchymes in the mesonephros, which can be distinguished by their specific expression of Gli1 and Amhr2, respectively. In the XX embryo, during sexual differentiation, the Wolffian duct degenerates. In this study, by tracking the fate of the Gli1+ Wolffian duct mesenchyme in the tamoxifen inducible genetic lineage tracing model Gli1-CreER; Rosa-Tomato, we demonstrate that the Wolffian duct mesenchyme does not regress along with the Wolffian duct epithelium in the XX embryo. Instead, the Wolffian duct mesenchyme is repurposed and becomes a unique mesenchymal population in the female reproductive tract. Profiling the transcriptomes and chromatin accessibilities of the Gli1+ Wolffian duct mesenchyme in XX and XY during sexual differentiation (E14.5 and E16.5) shows signaling pathways and transcriptional factors that potentially involves the female and male fate differentiation of the Gli1+ Wolffian duct mesenchyme. Comparing the transcriptomes between the Gli1+ Wolffian duct mesenchyme and the Amhr2+ Müllerian duct mesenchyme from the neonatal uterus reveals similarities and differences between the two mesenchymal populations in the female reproductive tract. Ex vivo ablation of the Wolffian duct mesenchyme stunted the growth of the fetal female reproductive tract. In summary, our discovery of the contribution of the Wolffian duct mesenchyme to the female reproductive tract provides new insights into our understanding of sexual differentiation of reproductive tracts.

Project description:Grb2 is a small SH2-SH3 adaptor molecule that interacts with activated tyrosine kinase receptors (RTKs), providing a crucial link towards downstream activation of pro-proliferative and pro-survival ERK and Akt signaling pathways. Ret and FGFR2, two RTKs that interact with Grb2, play important roles in ureteric branching and the proper establishment of the renal collecting duct system, but it is unclear whether Grb2 is required in this process. In this study, we selectively ablated a conditional floxed allele of the Grb2 gene within the ureteric epithelial lineage in mice and demonstrate that Grb2 signaling is essentially required for proper collecting duct development. Ureteric Grb2 deficiency results in perinatal lethality and severe renal hypodysplasia closely reminiscent of the rudimentary kidney phenotypes observed in Ret-null mutant mice. Grb2 loss attenuates ERK and Akt activation in the ureteric epithelia resulting in pronounced impairment of ureteric branching. Gene expression analysis reveals that Grb2 deficiency results in defective induction of genes implicated in both ureteric branching and reciprocal mesenchymal metanephric induction. Our findings therefore strongly indicate that Grb2 is a physiologically-relevant major RTK signaling relay partner that promotes renal branching morphogenesis and the proper development of the collecting duct system and the urogenital tract. Project description:Mitogen-Activated Protein 3 Kinase 1 (MAP3K1) is a dynamic signaling molecule with myriad cell-type specific functions not yet fully understood. Here we describe a role of MAP3K1 in the development of female reproductive tract (FRT). MAP3K1 kinase domain deficient females (Map3k1ΔKD) exhibit imperforate vagina, labor failure and sterility. The defects correspond to shunted Müllerian duct (MD), precursor of the FRT, in embryos, and contorted caudal vagina and abrogated vagina-urogenital sinus fusion in newborns. Although MAP3K1 acts through JNK and ERK to activate WNT in epithelial cells, it is dispensable for epithelial but crucial for mesenchymal WNT activity at the caudal embryonic MD in vivo. Moreover, conditional media derived from MAP3K1-compentent, but not -deficient, epithelial cells activate TCF-luciferase reporter in fibroblasts, suggesting MAP3K1 induces secreted activators from epithelial cells to activate WNT in fibroblasts. Correspondingly, the expression of Wnt7b is high in wild type, but low in Map3k1 knockout caudal MD epithelium and MAP3K1-deficient keratinocytes. Our data reveal a temporal-spatial paracrine MAP3K1-WNT crosstalk in the regulation of caudal MD elongation and FRT development.
