Project description:We have developed a new transgenic mouse strain, expressing a CyclinB1-GFP fusion reporter, which marks replicating cells in the S/G2/M phases of the cell cycle to isolate live replicating and quiescent cells from the liver. We used microarrays to compare the transcriptome of relicating and quiescent liver cells and reveal gene expression programs associated with cell proliferation in vivo Livers of three individual 3.5 week old, weaned CyclinB1-GFP mice were dissociated into single cells.Replicating (GFP-positive) and quiescent (GFP-negative) liver cells were FACS-sorted for RNA extraction and hybridization on Affymetrix microarrays.

Project description:Understanding the liver stem cells (LSCs) holds great promise for new insights into liver diseases and liver regeneration. By employing single-cell RNA-seq technology with weighted gene co-expression network analyses (WGCNA), we examined transcriptome features of distinct Krt19 lineage-tracing cell types isolated from Krt19CreERT; Rosa26R-GFP reporter mice, and identified gene expression signatures of quiescent and active adult LSCs, as well as their downstream lineage-restricted progenitors and terminally differentiation hepatocytes and cholangiocytes. Importantly, we discovered a novel cell surface LSC marker, CD63, as well as CD56, which distinguished active and quiescent LSCs. Furthermore, we confirmed that CD63+CD56- quiescent LSCs resided in Canals of Hering and peribiliary glands and that these cells could be activated by a combined action of VEGF and bFGF, and subsequently underwent terminal differentiation. These findings define an authentic adult liver stem cells compartment and highlight its contribution to liver homeostasis during pathophysiologic processes.

Project description:Hematopoietic stem cells (HSCs) are maintained in the quiescent state for protection from exhaustion by a number of self-renewal divisions. To understand the in vivo kinetics of quiescent HSCs, we analyzed the cell cycle of CD201+150+48-Lin-Kit+Sca-1+ cells after transplantation and during development and aging using fluorescent ubiquitination-based cell cycle indicator (Fucci) mice to distinguish HSCs at the G0, G1, and S/G2/M phases. The quiescent HSC population, representing a functional HSC pool, rapidly expanded by three weeks after transplantation and by three weeks of age in development and gradually accumulated in bone marrow with aging. Single-cell RNA-sequencing with flow cytometric index sorting suggested that high CD201 and Sca-1 expression levels and a low level of mitochondrial activity were associated with quiescent HSCs. A novel set of candidate quiescent genes in HSCs was also provided. This study implied that controlling quiescent HSCs is important for the in vivo expansion and maintenance of functional HSCs.
