Identification and functional analysis of SHISA2 overexpressed in high-grade prostate cancer cells.

ABSTRACT: We identified a novel molecular target and diagnostic biomarker, SHISA2, as an overexpressed gene in high-grade prostate cancer (PC) cells. To understand the association of SHISA2 overexpression with the aggressiveness of high-grade PC, we performed gene expression analysis using a cDNA microarray.

Project description:To identify molecules to serve as diagnostic markers for high-grade prostate cancer (PC) and targets for novel therapeutic drugs, we investigated the gene expression profiles of high-grade PCs using a cDNA microarray combined with laser microbeam microdissection. For this study, we collected 10 frozen specimens from high-grade PCs with high PSA levels and high Gleason score (GS) in clinically using prostatic needle biopsy. All needle biopsy specimens were at clinical stages T2 to T4 with or without N1 and M1 and their GS were 8-9. Moreover, all 10 patients had not received androgen ablation therapy. Simultaneously, normal prostate (NP) epithelial cells were also microdissected from five non-prostate cancer (BPH) patients. These NP cells from five males were used as a normal mixture control for our cDNA microarray analysis.

Project description:We performed copy number analysis of high-grade osteosarcoma samples. in order to detect osteosarcoma drivers, we integrated these data with genome-wide gene expression data. We performed two different methods - a non-paired and a paired integrative analysis. Copy number analysis was performed on 32 pre-treatment high-grade osteosarcoma diagnostic biopsies, of which 29 overlapped with the 84 samples used for gene expression analyses.

Long noncoding RNA LINC01111 suppresses pancreatic cancer aggressiveness by regulating DUSP1 expression via microRNA-3924

Project description:Dysfunction in long noncoding RNAs (lncRNAs) is reported to participate in the initiation and progression of human cancer; however, the biological functions and molecular mechanisms through which lncRNAs affect pancreatic cancer (PC) are largely unknown. Here, we report a novel lncRNA, LINC01111, that is clearly downregulated in PC tissues and plasma of PC patients and acts as a tumor suppressor. We found that the LINC01111 level was negatively correlated with the TNM stage but positively correlated with the survival of PC patients. The overexpression of LINC01111 significantly inhibited cell proliferation, the cell cycle, and cell invasion and migration in vitro, as well as tumorigenesis and metastasis in vivo. Conversely, the knockdown of LINC01111 enhanced cell proliferation, the cell cycle, and cell invasion and migration in vitro, as well as tumorigenesis and metastasis in vivo. Furthermore, we found that high expression levels of LINC01111 upregulated DUSP1 levels by sequestering miR-3924, resulting in the blockage of SAPK phosphorylation and the inactivation of the SAPK/JNK signaling pathway in PC cells and thus inhibiting PC aggressiveness. Overall, these data reveal that LINC01111 is a potential diagnostic biomarker for PC patients, and the newly identified LINC01111/miR-3924/DUSP1 axis can modulate PC initiation and development.

Histology-Resolved Proteomics Identifies Tumor and Stromal Proteome Profiles Associated with Low- and High-Grade Prostate Cancer

Project description:Prostate cancer is one of the most frequently diagnosed cancers in men. Prostate tumor staging and disease aggressiveness are evaluated based on the Gleason scoring system, which is further used to direct clinical intervention. The Gleason scoring system provides an estimate of tumor aggressiveness through quantitation of the serum level of prostate specific antigen (PSA) and histologic assessment of Grade Group, determined by the Gleason Grade of the tumor specimen. To improve our understanding of the proteomic characteristics differentiating low- versus high-grade prostate cancer tumors, we performed a deep proteomic characterization of laser microdissected epithelial and stromal subpopulations from surgically resected tissue specimens from patients with Gleason 6 (n=23 specimens from n=15 patients) and Gleason 9 (n=15 specimens from n=15 patients) prostate cancer via quantitative high-resolution liquid chromatography-tandem mass spectrometry analysis.

Genome-wide gene expression analysis was performed on 84 pre-treatment high-grade osteosarcoma diagnostic biopsies, of which 29 overlapped with the 32 samples used for copy number analysis. Two different sets of control samples were used for comparison: osteoblasts (n=3) and mesenchymal stem cells (n=12, GEO accession number GSE28974).

RNA sequencing analysis of JAG1 overexpressed Ink4a-/- murine astrocytes

Project description:Notch signaling is a signaling pathway related to development and homeostasis of tissue. Jagged 1 (JAG1), which is one of the ligand of NOTCH, is highly expressed in tumor and induce proliferation, survival and tumor aggressiveness of cancer cells. This study demonstrate the biological function of JAG1 in tumor progression. To elucidate the role of JAG1, JAG1 was overexpressed in murine Ink4a-/- astrocytes. Our results provide the clue of molecular mechanism in tumor aggressiveness.
