ABSTRACT: Preterm birth is an important unsolved clinical problem. Despite advanced treatments, infants who survive prematurity remain at increased risk for permanent disabilities. In approximately one-third of cases, prematurity is related to infection and/or inflammation, which renders hostile the normally receptive intrauterine environment. Proinflammatory cytokines provoke up-regulation of genes that promote uterine contractions. Using monolayer cultures of human decidual cells as a model, we profiled the global pattern of gene expression in response to cytokine challenge.

Using monolayer cultures of human decidual cells as a model, we profiled the global pattern of gene expression in response to cytokine challenge. Decidualized human endometrial stromal cells were treated with 1 ng/ml of interleukin-1-beta for 6 hours, while control cells were treated with 0.1% PBS containing 0.1% bovine serum albumin (vehicle). The cells were subjected to RNA extraction, quantification, and evaluation, followed by hybridization on Affymetrix Human Gene 2.0 ST Arrays (Platform GPL16686). Three biological replicates were used (6 total samples).

Proteomic profiling of human decidual immune proteins during Toxoplasma gondii infection

Project description:A Toxoplasma gondii infection during pregnancy can result in spontaneous abortion, preterm labor, or congenital fetal defects. The decidual immune system plays a critical role in regulating the immune micro-environment and in the induction of immune tolerance. To better understand the factors that mediate the decidual immune response associated with the T. gondii infection, a large-scale study employing TMT proteomics was conducted to characterize the differential decidual immune proteomes from infected and uninfected human decidual immune cells samples. The decidual immune cells from 105 human voluntary abortion tissues were purified, and of the 5510 unique proteins identified, 181 proteins were found to be differentially abundant (>1.2-fold cutoff, P＜0.05) in the T. gondii-infected decidual immune cells. 11 proteins of 181 differentially expressed proteins associated with trophoblast invasion, placental development, intrauterine fetal growth, and immune tolerance were verified using a quantitative real-time polymerase chain reaction and western blotting. This systematic research identified a broad range of immune factors in human decidual immune cells, shedding a new insight into the decidual immune molecular mechanism for abnormal pregnancy outcomes associated with T. gondii infection.

Reconstruction of the decidual pathway in human endometrial cells using single-cell RNA-Seq

Project description:The fate of the human endometrium is determined during the mid-luteal window of implantation, coinciding with differentiation of endometrial stromal cells (EnSCs) into specialized decidual cells. Upon embryo implantation, differentiating EnSCs transform the endometrium into the decidua of pregnancy; whereas falling progesterone levels in the absence of pregnancy lead to tissue breakdown and menstruation. We used single-cell RNA sequencing to map the transcriptomic changes in primary EnSCs along a decidual time-course and in response to withdrawal of differentiation signals. We demonstrate that decidual transformation starts with a precipitous transcriptional response, which is followed by synchronous transition of EnSCs through intermediate states before emerging as divergent subpopulations, representing decidual cells and senescent decidual cells. Single-cell analysis of timed luteal phase biopsies identified multiple endometrial epithelial subpopulations and immune cells, most prominently uterine NK cells. In the stroma, transition from receptive to post-receptive endometrial state was marked by progression of EnSCs along diverging transcriptional trajectories, involving genes with conserved branching dynamics in vivo and in vitro. Our findings indicate that specification of EnSCs into distinct decidual subpopulations underpins endometrial fate decisions during the window of implantation.

Placental exosomes polarize human monocyte-derived macrophages to a decidual M2-like macrophage phenotype

Project description:Macrophages are abundant in uterine mucosa during the peri-implantation phase and early pregnancy. Decidual macrophages display dynamic changes alone with pregnancy progress: During the peri-implantation phase, macrophages displayed a pro-inflammatory phenotype which facilitates embryo implantation. While, In the late firster trimester and second trimester, decidual macrophages are anti-proinflammatory which are helpful to pregnancy maintenance. Alterations in the ratio of pro-inflammatory/anti-inflammatory decidual macrophages lead to abortion, preeclampsia, and preterm birth. Placenta-derived exosomes (pEXO) are critical in the immune cell modulation such as T cell apoptosis, NK activities, and T regulatory (Treg) differentiation. However, it is unknown whether placenta-derived exosomes contribute to decidual macrophage polarization during early pregnancy. Here we report that exosomes from the placenta explant stimulate M2 macrophage polarization via exosomal miRNA-30d-5p. Mechanistically, miRNA-30d-5p polarized macrophages to M2 phenotype by inhibiting HDAC9 expression. Furthermore, the conditioned medium of pEXO-treated macrophages promoted trophoblast migration and invasion. By contrast, conditioned medium impaired the ability of endothelial cell tube formation. However, pEXO-treated macrophages have no impact on T cell proliferation. Together, we demonstrated that pEXO promoted trophoblast migration and invasion, endothelial cell migration, and attenuation of endothelial cell tube formation by polarizing macrophage to decidua-like macrophage.

Mechanisms of Chorioamnionitis-associated Preterm Birth: Interleukin 1 β inhibits Progesterone Receptor Expression in Decidual Cells

Project description:In chorioamnionitis (CAM), a major cause of preterm birth (PTB), maternal-fetal inflammatory responses in the decidua and amnio-chorion cause the release of cytokines that elicit cervical ripening, fetal membrane rupture and myometrial activation. We posited that this inflammatory milieu can trigger PTB via inhibited progesterone receptor (PR) expression and increased decidual prostaglandin (PG) production. We found significantly lower decidual cell PR levels in CAM-complicated PTB using immunohistochemistry. Decidual cells (DCs) treated with IL-1β displayed decreased PR expression and significantly increased PGE2 and PGF2α production and COX2 expression. While addition of PGF2α to DC cultures was also found to suppress PR expression, the COX inhibitor, indomethacin, did not reverse IL-1β suppression of PR expression in DC cultures. Although IL-1β treatment activated NF-B, ERK1/2 and p38 MAPK signaling cascades in DCs, only inhibition of ERK1/2 MAPK signaling completely reversed IL-1β suppressed PR levels. These findings suggest that CAM-associated PTB is induced at least in part by IL-1β-mediated functional progesterone withdrawal.

Project description:Premature birth continues to be a challenging pregnancy complication, and a body of literature indicates that inflammation can contribute to premature delivery by converting a receptive uterine environment to a hostile one. Cytokines have been demonstrated to provoke up-regulation of inflammatory genes (e.g. interleukin-1, 6, and 8, tumor necrosis factor-alpha, cyclooxygenase-2, and microsomal prostaglandin E synthase-1). Using monolayer cultures of human amnion mesenchymal cells (AMCs) as a model, we aimed to detail the global programme of gene expression that occurs in response to cytokine challenge. Human amnion mesenchymal cells were challenged with 10 ng/ml of interleukin-1-beta for 1 hour (1h) or 8 hours (8h) in serum-free media, while control cells were treated with serum free medium only (veh). The cells (veh, 1h and 8h) were subjected to RNA extraction, quantification, and evaluation, followed by hybridization on Affymetrix GeneChip® Human Genome U133A 2.0 Arrays (Platform GPL571). Experiments were repeated three times for a total of nine samples used for microarray analysis. Three biological replicates were used to ensure that any results were not biased by day-to-day variation in RNA extraction.

miRNA expression in tracheal aspirates

Project description:Extreme preterm infants are a growing population in neonatal intensive care units who carry a high mortality and morbidity. Multiple factors play a role in preterm birth, resulting in major impact on organogenesis leading to complications including bronchopulmonary dysplasia (BPD). The goal of this study was to identify biomarker signatures associated with prematurity and BPD. We analyzed miRNA and mRNA profiles in tracheal aspirates (TAs) from 51 infants receiving invasive mechanical ventilation. 25 infants were extremely preterm and diagnosed with BPD, and 26 were term babies receiving invasive mechanical ventilation for elective procedures. We found specific mRNA-miRNA signatures in TAs that may serve as biomarkers for BPD pathogenesis, a consequence of extreme prematurity.
