A highly clonogenic subset of adult mouse biliary duct cells

ABSTRACT: Our goal is to find the heterogeneity of mouse liver ductal cells in the wild type mouse liver. We separated the MIC1-1C3+ mouse liver ductal cells into two populations with the cell surface marker: ST14. We found that ST14hi1C3+ cells have higher colonal geneity comparing to the ST14lo1C3+ population.

Prospective isolation of a bipotential clonogenic liver progenitor cell in adult mice

Project description:The molecular identification of adult hepatic stem/progenitor cells has been hampered by the lack of truly specific markers. To isolate putative adult liver progenitor cells, we used cell surface-marking antibodies, including MIC1-1C3, to isolate subpopulations of liver cells from normal adult mice or those undergoing an oval cell response and tested their capacity to form bilineage colonies in vitro. Robust clonogenic activity was found to be restricted to a subset of biliary duct cells antigenically defined as CD45–/CD11b–/CD31–/MIC1-1C3+/CD133+/CD26–, at a frequency of one of 34 or one of 25 in normal or oval cell injury livers, respectively. Gene expression analyses revealed that Sox9 was expressed exclusively in this subpopulation of normal liver cells and was highly enriched relative to other cell fractions in injured livers. In vivo lineage tracing using Sox9creERT2-R26RYFP mice revealed that the cells that proliferate during progenitor-driven liver regeneration are progeny of Sox9-expressing precursors. A comprehensive array-based comparison of gene expression in progenitor-enriched and progenitor-depleted cells from both normal and DDC (3,5-diethoxycarbonyl-1,4-dihydrocollidine or diethyl1,4-dihydro-2,4,6-trimethyl- 3,5-pyridinedicarboxylate)-treated livers revealed new potential regulators of liver progenitors.

Human biliary epithelial cells from discarded donor livers rescue bile duct structure and function in a mouse model of biliary disease

Project description:Biliary diseases can cause inflammation, fibrosis, bile duct destruction and eventually liver failure. There are no curative treatments for biliary disease except for liver transplantation. New therapies are urgently required. We have purified human Biliary Epithelial Cells (hBEC) from discarded human livers. hBECs were tested as a cell therapy in a mouse model of biliary disease where the conditional deletion of Mdm2 in cholangiocytes causes senescence, biliary strictures and fibrosis. hBEC are expandable, phenotypically stable and help restore biliary structure and function, highlighting their regenerative capacity and a potential alternative to liver transplantation for biliary disease.

Liver-restricted deletion of the biliary atresia candidate gene Pkd1l1 causes bile duct dysmorphogenesis and ciliopathy

Project description:A recent multicenter genetic exploration of the biliary atresia splenic malformation (BASM) syndrome identified mutations in the ciliary gene PKD1L1 as candidate etiologic contributors. We hypothesized that deletion of Pkd1l1 in hepatoblasts would provide a mouse model of the developmental cholangiopathy of BA. We then performed gene expression profiling analysis using data obtained from total RNA-seq, isolated from the livers of Pkd1l1Fl/Fl (Fl/Fl) and Pkd1l1Fl/Fl x AFP-CRE mice (Pkd1l1ΔExon8/ΔExon8 Liver knockout ; LKO) at 7 weeks.

Nuclear GSK-3β and Oncogenic KRas Promote Aqp5+ Pancreatic Duct Stem Cell Expansion

Project description:Intraductal papillary mucinous neoplasm (IPMN) represents the most commonly diagnosed precursor lesion of pancreatic ductal adenocarcinoma (PDA), however the cell-of-origin remains unclear. Herein, we show that pancreas-specific activation in mice of nuclear-targeted glycogen synthase kinase-3β and oncogenic KRasG12D leads to the loss of acinar cells, the expansion of ductal cells, and the rapid development of IPMN with low-grade dysplasia. RNA-sequencing identified the expression of several ductal stem cell lineage genes including the water channel AQP5. The Aqp5+ pancreatic ductal cell pool was proliferative, phenotypically distinct from mature pancreatic ductal cells, and deletion of AQP5 limited IPMN development. Significantly, Aqp5 is highly expressed in human IPMN along with GSK-3b suggesting that human preneoplastic lesions likely arise from the expansion of an Aqp5+ pancreatic ductal stem cell. Altogether, these data identify Aqp5+ ductal cells in the mouse and human pancreas as the likely cell-of-origin for IPMN.

NGFR-marked basal duct progenitors drive ductal-acinar regeneration in injured salivary glands

Project description:Severe salivary gland injury causes long-term structural and functional impairment, yet the epithelial populations associated with repair remain incompletely defined. Here, we identify NGFR as a surface marker of a basal duct-associated epithelial population with organoid-forming capacity in the human salivary gland and show that Ngfr marks a conserved injury-responsive epithelial population in the mouse salivary gland. Single-cell RNA sequencing of human salivary gland tissue revealed that NGFR was enriched in a restricted basal duct subpopulation distinct from differentiated epithelial and myoepithelial compartments. NGFR-positive cells exhibited a progenitor-like transcriptional profile and were enriched at early pseudotime positions in inferred ductal-acinar differentiation trajectories. Functionally, NGFR-positive cells isolated from human tissue and organoids showed enhanced organoid-forming capacity, and NGFR-enriched organoids engrafted into injured mouse salivary glands. In mouse salivary glands, Ngfr marked a rare epithelial population whose isolated Ngfr-positive fraction showed enriched organoid-forming activity and was detected in injury-associated ductal regions after duct ligation and local inflammatory injury. Lineage tracing using Ngfr-CreERT2; Rosa26-tdTomato mice further showed that Ngfr-lineage cells contribute to ductal and acinar epithelial compartments during post-injury regeneration. Together, these findings define NGFR in human and Ngfr in mouse as conserved markers of an injury-responsive basal duct epithelial population that can be prospectively isolated and contributes to ductal and acinar epithelial compartments during post-injury salivary gland regeneration.
