DNA methylation profiling of vascular maturation

ABSTRACT: Endothelial cells line the inside of all blood vessels forming a single layer of quiescent, non-proliferating cells. While the mechanisms of sprouting angiogenesis, network formation and vascular remodelling are molecularly unravelled in increasing detail, the molecular mechanisms of vascular maturation are still poorly understood. We performed whole-genome bisulfite sequencing on endothelial cells isolated from infant and adult mice to establish the epigenetic landscape of vascular maturation. Overall design: Tagmentation-based Whole-Genome Bisulfite Sequencing of infant and young adult lung endothelial cells. Three independent biological replicates each were analyzed.

INSTRUMENT(S): Illumina HiSeq 2000 (Mus musculus)

Maintenance of a quiescent and organotypically-differentiated layer of blood vessel-lining endothelial cells (EC) is vital for human health. Yet, the molecular mechanisms of vascular quiescence remain largely elusive. Here we identify the genome-wide transcriptomic program controlling the acquisition of quiescence by comparing lung EC of infant and adult mice, revealing a prominent regulation of TGFß family members. These transcriptomic changes are distinctly accompanied by epigenetic modificati  ...[more]
