MICRORNA EXPRESSİON PATTERNS OF CD8+ T CELLS IN ACUTE AND CHRONIC BRUCELLOSIS

Evaluation of immunological factors involved in initiation of chronic brucellosis in CD4+ T cells using miRNA array

Project description:Brucellosis is a serious infectious disease and continues to be an important cause of morbidity. It can be seen almost anywhere in the world and at any age. Acute phase heals or becomes chronic form. Infection of 10-30% of patients becomes chronic, despite early diagnosis and treatment. Although our knowledge about Brucella virulence factors and the host response increase rapidly, how they can hidden from the immune system and cause chronic disease are still unknown. We aimed to investigate the immunological factors which belong to CD4+ T cells and their roles in the transition of brucellosis from acute to chronic infection. Using miRNA microarray, more than 2000 miRNAs were screened in CD4 + T cells of patients with acute or chronic brucellosis and healthy controls that were sorted from peripheral blood with flow cytometry and validated by RT-qPCR. Findings were evaluated using GeneSpring GX (Agilent) 13.0 software and KEGG pathway analysis. In comparison to acute cases, expression levels of 28 miRNAs were significantly altered in chronic cases. All of miRNAs were up-regulated in chronic cases in comparison with the control group. Apart from a miRNA (miR-4649-3p), 27 miRNAs were not expressed in the acute cases (p <0.05, fold change> 2). These miRNAs have the potential to be markers for chronic cases. The differential expressed miRNAs and their predicted target genes that involved in previously MAPK signaling pathway, regulation of actin cytoskeleton, endocytosis, and protein processing in endoplasmic reticulum indicating their potential roles in chronic brucellosis and its progression. It is the first study of miRNA expression analyzes of human CD4+ T cells to clarify the mechanism of inveteracy in brucellosis.

Project description:In an attempt to identify novel markers and immunologic targets in leukemic stem cells (LSC) in acute myeloid leukemia (AML) and chronic myeloid leukemia (CML), we screened samples of patients with AML, CML, and controls for expression of cell surface antigens on CD34+/CD38− and CD34+/CD38+ cells by multi-color flow cytometry. In addition, we examined mRNA expression profiles in highly purified CD34+/CD38− and CD34+/CD38+ cells by gene array- and qPCR analyses. Aberrantly expressed markers were identified in all patient cohorts examined.

Unraveling Potential Biomarkers for Acute and Chronic Brucellosis Through Proteomic and Bioinformatic Approaches

Project description:This study aimed to identify potential biomarkers associated with acute and chronic brucellosis by employing advanced proteomic and bioinformatic methodologies. Blood samples from individuals with acute and chronic brucellosis and healthy controls were analyzed. Differentially expressed proteins (DEPs) were identified using differential expression analysis and weighted gene co-expression network analysis (WGCNA). A total of 25 DEPs were identified in all three pairwise comparisons, and 20 hub proteins were identified in the WGCNA analysis. Nine proteins overlapped between the two analyses, highlighting their potential importance in brucellosis. A random forest model based on these nine proteins demonstrated good classification performance for the three groups. Enrichment analysis revealed that these proteins are involved in various biological processes, including inflammation, coagulation, extracellular matrix regulation, and immune response. The results offer valuable insights into potential therapeutic targets or diagnostic biomarkers for brucellosis.

Project description:In a murine, adoptive transfer model of chronic lymphocytic leukemia we injected OT-I cells followed by infection with mCMV-OVA. On day 7 post-infection, OT-I cells from the spleen were FACS sorted and studied with ATAC sequencing and RNA sequencing. In additition to flow cytometry data demonstrating a skewing towards short-lived effector cells and a lower proportion of memory-precursor cells of OT-I cells from CLL mice compared to WT, we also found effector and memory genes epigenetically and transcriptionally altered.

Transcription profiling of mouse naive, effector, memory and exhausted virus-specific CD8 T cells after chronic viral infection

Project description:CD8 T cells normally differentiate from resting naÃ¯ve T cells into function effector and then memory CD8 T cells following acute infections. During chronic viral infections, however, virus-specific CD8 T cells often become exhausted. We used microarrays to examine the gene expression differences between naive, effector, memory and exhausted virus-specific CD8 T cells following lymphocytic choriomeningitis virus infection. Experiment Overall Design: Three or four independent samples were sorted by flow cytometry for each cell type (naive, effector, memory and exhausted) virus-specific CD8 T cells. RNA was extracted and hybridized to Affymetrix microarrays.

Crizotinib, Tyrosine Kinase Inhibitors, Create an Antibacterial Active against Gram-positive Bacteria by Targeting the CTP Synthetase PyrG

Project description:Abstract The infection of drug resistant bacteria seriously threats to public health, which makes it more urgent to find novel antibacterial compounds. Compared with traditional development approaches, drug repurposing provides a faster and more effective strategy to find new antimicrobial agents. Here, we screened an FDA-approved small-molecule library upon S. aureus, and identified crizotinib as an antimicrobial agent. We confirmed the antibacterial activities of crizotinib in vitro and in vivo by MIC, growth curve, SEM and mice experiment. At the same time, crizotinib also showed low tendency to develop resistance. Mechanistically, quantitative proteomics, bioinformatics analyses, qRT-PCR and flow cytometry biochemical validation experience confirmed that crizotinib exerted its antibacterial effects by interfering pyrimidine metabolism and disrupting DNA synthesis eventually. Furthermore, our data from drug affinity responsive target stability, bio-layer interferometry and a series of functional assays demonstrated that crizotinib could target PyrG directly in pyrimidine metabolism pathway. Taken together, our results indicated that crizotinib could be a potential antimicrobial agent to treat Gram-positive bacterial infections in the future.
