MiRNAs in flowers of Viburnum macrocephalum f. keteleeri

ABSTRACT: High-throughput small RNA sequencing were performed to identify a large number of miRNAs and their targets in fertile and sterile flowers of V. macrocephalum f. keteleeri for the first time. We ascertained that the regulatory networks of the miRNAs are involved in many different primary biological processes based on potential target designations. Our study is the first to provide useful information for uncovering the regulatory networks of miRNAs in V. macrocephalum f. keteleeri flowers.

Project description:Dominant genic male sterility (DGMS) is a valuable system for hybrid seed production and improvement of plant populations, but the molecular mechanism of DGMS is not well understood. To facilitate the analysis of mechanism underlying DGMS, we compared gene expression profiles between fertile and sterile flowers of two pair of Brassica napus isogenic lines: J10AB and J03AB which only differ in sterility-related locus and fertility restore-related locus respectively by using Arabidopsis thaliana ATH-1 microarray. Gene expression profiling revealed that 360 genes differed in expression between the two lines and the number of differently expressed genes increased with progressing developmental stages. Cluster analysis result indicates that most important gene expression changes occur from PMC (anther less than 1mm) to uninucleate stage (anther in 3mm). There are 182 differential expressed genes detected between fertile plants and sterile plants at same stage. Some important pathways related to fertility control by KEGG analysis, such as glycolysis/gluconeogenesis pathway, flavonoid biosynthesis pathway and fatty acid metabolism pathway. Another abortion usually associated with suppression of gene expression of these pathways. Experiment Overall Design: The Homozygous DGMS two-type line J10AB was compared with heterozygous two-type line J03AB at three different stages of development: tetrad stage, uninucleate stage and trinucleate stage (Flower buds <1, 1-3 and >3mm, respectively) which were confirmed by cytology observation. All these flower buds were collected by hand. For each stage, two biological repetitions and two experimental repetitions were used, resulting in four hybridizations per developmental stage. Gene expression was compared between lines at different stages of development, as well as between developmental stages of fertile plants and sterile plants of each line.

Transcriptional profiling of Persea americana flowers

Project description:The debate on the origin and evolution of flowers has recently entered the field of developmental genetics, with focus on the design of the ancestral floral regulatory program. Flowers can differ dramatically among angiosperm lineages, but in general, sterile perianth organs surrounding stamens (male reproductive organs) and carpels (female reproductive organs) constitute the basic floral structure. However, the basal angiosperm lineages exhibit spectacular diversity in the number, arrangement, and structure, of floral organs, while the evolutionarily derived monocot and eudicot lineages share a far more uniform floral ground plan. As such, regulatory mechanisms underlying the archetypal floral plan, for instance that of the eudicot genetic model Arabidopsis thaliana, are unlikely to apply to the original flowers. Here we show that broadly overlapping transcriptional programs characterise the floral transcriptome of the basal angiosperm Persea americana (avocado), while floral gene expression domains are typically organ-specific in Arabidopsis. Our findings extend the “fading borders” model for basal angiosperms from organ identity genes to the downstream floral transcriptome, and suggest that the combinatorial mechanism for organ identity may not operate in basal angiosperms as it does in Arabidopsis. Furthermore, fading expression of components of the stamen transcriptome in central and peripheral regions of Persea flowers resembles the developmental program of the hypothesized gymnosperm “floral progenitor”. Accordingly, in contrast to the canalized organ-specific regulatory apparatus of Arabidopsis, floral development may have been originally regulated by overlapping transcriptional cascades with fading gradients of influence from focal to bordering organs.

Microarray analysis of male strobili in fertile and sterile male strobili of Cryptomeria japonica

Project description:Investigation of comprehensive gene expression level changes in male strobili of sterile mutant, compared to the fertile plant. We designed a new custum array derived from ESTs of Cryptomeria japonica for this study. Male strobili of C. japonica were collected from Toyama 1 half-sib F2 pedigree. Toyama 1 male-sterile mutant was first reported in Saito, M. , H. Taira, and Y. Furuta (Journal of Forest Research 3(3):167-173). Microarray studies using both individual samples and pooled samples were performed. Total RNA was extracted from male strobili at four developmental stages derived from each seven fertile and male-sterile trees as individual samples. Pooled RNA samples were prepared from four to eight different fertile and sterile plants at three developmental stages.

Transcriptome profiling of differentially expressed genes from floral buds and flowers of male sterile and fertile lines in watermelon

Project description:Male sterility is important mechanism in watermelon for production of hybrid seed. While some fruit development related studies were widely performed in watermelon, there are no reports of profiling gene expression in floral organs of watermelon. RNA-seq analysis was performed in order to identify male sterility related genes from two different groups of watermelon (genetic male-sterile (GMS) DAH3615-MS line and male-fertile DAH3615 line, respectively) to identify the differentially expressed genes (DEGs).
