Project description:Here we compared the expression of an engineered kidney tissue, created by recombining an in vitro budded Wolffian duct with fresh E13 metanephric mesenchyme, with that of three in vivo rat embryonic kidney timepoints (E13, E18, and week 4) Experiment Overall Design: The global gene expression of recombined tissue (non-branched in vitro-formed UBs and MM) was analyzed and compared to the gene expression of early, late, and post developmental kidneys in order to determine whether normal developmental pathways were being followed. Genes with at least a 3-fold difference in expression between any of the 4 conditions (E13, E18, Wk4, recombined tissue) were analyzed and grouped into one of ten expression patterns. The expression of each group of genes was analyzed in the recombined tissue to compare the recombined tissue gene expression levels to the three in vivo time points.

Transcription profiling of rat kidney progenitor tissue from embryo at E13

Project description:In this study we compared genes expressed in the unbudded portion of the Wolffian duct with the isolated ureteric bud to find genes novel to early kidney development. We used the Affymetrix Rat Genome 230 2.0 Array to compare the unbudded tissues with the budded samples. We used microarrays to find novel genes regulating early kidney formation. Experiment Overall Design: Isolated ureteric buds (UB) were separated from the embryonic kidney after a light trypsinization step. Approximately 100 UBs were pooled together into each replicate. Approximately 100 isolated Wolffian ducts were dissected from E13 rat embryoes and pooled into each sample. Approximately 50 Wolffian ducts with attached mesodermal cells were pooled into each replicate.

Project description:E11.5 metanephric mesenchyme and ureteric bud were dissected from the E11.5 kidney rudiment using fine manual microdissection (ureteric bud only) or both fine manual microdissection and laser capture microdissection (metanephric mesenchyme) to define the gene expression profiles of these structures. Additionally, HoxA11, HoxD11 compound null E11.5 metanephric mesenchyme was obtained through laser capture microdissection allowing analysis of possible Hox targets in kidney development. Targets from multiple biological replicates of each were generated and the expression profiles were determined using Affymetrix MOE430_v2 arrays. Using microdissection techniques, ureteric bud and metanephric mesenchyme were dissected from E11.5 kidney rudiments allowing the identificated genes specifically regulated in either structure. In addition, Hoxa11, Hoxd11 compound null E11.5 metanephric mesenchyme were normalized to wild type embryonic controls allowing the identification of potential Hox targets in normal kidney development. Each structure/genotype were represented in biological (seperate embryo) replicate.

Gene expression change induced by bisphenol A in mouse urogenital sinus

Project description:Bisphenol A (BPA), an endocrine-disrupting chemical (EDC), is a well-known, ubiquitous estrogenic chemical. To investigate the effects of fetal exposure to low-dose BPA on the development of the prostate, we first examined the alterations of in situ sex steroid hormonal environment in the mouse urogenital sinus (UGS). Next, to investigate the BPA-specific gene alterations related to increases of the E2 levels and aromatase activity, we performed comprehensive gene expression analysis using Affymetrix GeneChip in the BPA-treated or DES-treated male UGS at embryonic day 17th and postnatal day 1st. Pregnant female C57BL/6 mice were exposed to BPA (20 μg/kg/day) or synthetic estrogen Diethylstilbestrol (DES: 0.2 μg/kg/day), which were dissolved in tocopherol-stripped corn oil, on embryonic day 13 (E13) to E16. Between E17 and postnatal day 1 (P1), all animals were terminated by an overdose of isoflurane followed by cervical dislocation. Fetuses were collected at E17, E18, P0, and P1. The bladder and urethra were removed and dissected to isolate UGS and collected in RNA later. To isolate pure UGS, other tissues such as the bladder, urethra, Wolffian duct (WD), seminal vesicle (SV), and Mullerian duct (MD) were removed from both the male and female urogenital tracts. The histopathology of the mouse UGS was then examined by hematoxylin and eosin staining. Total RNA was extracted using the Qiagen mini RNA Easy kit. Each RNAs were linearly amplified and hybridized to Affymetrix GeneChip.
