MiRNA expression data from exosome subpopulations of OVCAR3 and HaCaT cells

ABSTRACT: Exosome subpopulations were separated based on differential affinity of exosome subpopulations to hydroxyapatite. The exosome profiles showed that exosome subpopulations derived from ovarian cancer cells are more heterogeneous than normal cell secreted ones. Especially, cancer cell-secreted exosomes have stronger affinity to the sorbent as compared to normal cell-released ones. We used microarrays to detail the miRNA expression data from exosome subpopulations of OVCAR3 and HaCaT cells.

Protein dataset of immortalized HaCaT cells p53 knock down HaCaT keratinocytes

Project description:The p53 protein is encoded by TP53 gene and plays the key role in significant number of cellular processes including proliferation, apoptosis and regulation of many stress response pathways. P53 acts like a direct transcription activator of numerous genes regulating cell cycle arrest, DNA repair, growth inhibition and many others (Mollereau and Ma, 2014). The canonical biological function of p53 is maintaining genome integrity via elimination of damaged or exposed to genotoxic stress cells. Immortalized HaCaT cells are widely used for keratinocyte research, since they maintain stable keratinocyte phenotype, have nearly unlimited proliferative potential, do not require specific growth and differentiation factors (Colombo et al., 2017). Also, HaCaT cells produce typical differentiation markers such as cytokeratins K14 and K10, involucrin (Colombo et al., 2017) and respond to keratinocyte differentiation stimuli. Taking together, HaCaT cells have similar to normal human keratinocytes (NHK) properties, however, as many of spontaneously immortalized cell lines HaCaT cells bear two mutant p53 alleles - R282Q and H179Y (Lehman et al., 1993). Mutp53 in HaCaT has an increased affinity to other p53 family members (p63, p73), which significantly expands p53 properties. Moreover, mutp53 indirectly affects specific target genes via protein-protein interactions with other transcription factors (NF-Y, E2F1, NF-KB) or by tethering p63 to new promotor locations. For more detailed investigation of mutp53 impact on various processes in HaCaT cells we performed a shRNA mediated knockdown of mutp53. For generation of stable TP53 knockdown we employed plasmid vector pLKO-p53-shRNA-941 (Addgene # #25637) followed by puromycin selection of transduced cells. Here we present proteomic dataset obtained from wild type HaCaT cells and p53 knock down HaCaT keratinocytes.

RNASET2-silenced and control OVCAR3 cell clones injected into nude mice.

Project description:To investigate the molecular pathways underlying RNASET2-mediated tumor suppression in vivo, we investigated the gene expression profile of tumor samples obtained from control and RNASET2-silenced OVCAR3 xenografts, respectively. In order to evaluate the contribution of human cancer cells and the murine host stroma to RNASET2-mediated tumorigenesis responses, total RNA extracted from tumor xenografts derived from six independent OVCAR3 cell clones (three parental and three RNASET2-silenced clones) was fluorescently labeled and hybridized to human Agilent whole-genome microarrays. Control and RNASET2-silenced OVCAR3 were injected subcutaneously into nude mice. After 39 days mice were sacrified, tumors were extracted and total RNA purified.

lncRNA and mRNA expression signatures in OVCAR3 cells: control vs. miR-200a transfected

Project description:To further development of the effects of miR-200a in ovarian cancer OVCAR3 cells，we have employed lncRNA and mRNA microarray as a discovery platform to identify lncRNA and mRNA expression in miR-200a overexpressing ovarian cancer cells. OVCAR3 cells were transfected with lentiviral vector with eGFP, encoding miR-200a and negative control vector (LV- miR-200a and LV-CON,) by using polybrene. The dysregulation of miR-200a was confirmed by using RT-PCR. RNA was extracted and detected by a lncRNA and mRNA microarray in LV-miR-200a and LV-CON OVCAR3 cells. The different expression of lncRNA and mRNA in LV-miR-200a and LV-CON OVCAR3 cells was analyzed to explore the mechanism that miR-200a affect ovarain cancer cells.

Cisplatin-Induced Changes in Proteomic Profiles of Ovarian Cancer Cell and HaCaT Secretomes

Project description:Exogenous signals from drug-stressed cancer cells promote acquisition of a more aggressive phenotype of neighboring tumor cells. To identify extracellular signaling molecules that potentially contribute to the development of therapy resistance, we examined how cancer cell secretome changes in response to chemotherapy. We performed LC-MS/MS analysis of secretomes from several ovarian cancer cell lines representing different tumor subtypes: serous ovarian cancer cell line (OVCAR3), ovarian cystadenocarcinoma cell line (MESOV) and clear cell ovarian cancer primary culture isolated from ascites (cells 26) before and 48 hours after cisplatin treatment. In addition, we studied secretomes from keratinocyte-derived epithelial cell line HaCaT as non-cancerous “normal” cells. Functional annotation of proteins which secretion increased after cisplatin showed that these proteins were mainly associated with cluster of spliceosome.

Identification progenitor Keratinocyte Subpopulations in HaCaT via high throughput sequencing

Project description:Epidermal progenitor cells rely on tightly regulated transcriptional and epigenetic programs to preserve cellular plasticity and prevent premature differentiation; however, these regulatory mechanisms remain incompletely characterized. In this study, we investigated the transcriptomic architecture of progenitor keratinocyte subpopulations isolated from HaCaT cells using fluorescence-activated cell sorting and bulk RNA sequencing. Differential expression analysis revealed distinct protein-coding and long non-coding RNA (lncRNA) profiles between subpopulations enriched for progenitor-associated markers and more differentiated cells. Enrichment analyses providing a transcriptomic framework for exploring epigenetic regulation of keratinocyte progenitor identity.
