The 11-hydroxy-jasmonic acid pathway expands jasmonate catabolic repertoire for stress-specific hormone attenuation in Arabidopsis

Project description:Jasmonates are plant hormones undergoing complex metabolic conversions, generating organ- or stimulus-specific signatures. Several jasmonates result from catabolism of the master regulator jasmonoyl-isoleucine (JA-Ile) that controls most jasmonate responses upon developmental or environmental cues. Hydroxylation of the precursor jasmonic acid by Jasmonic Acid Oxidases (JAO) holds a peculiar regulatory function, by attenuating basal JA-Ile formation and action, and promoting growth. Here we re-investigated the nature and origins of hydroxy-JAs and their derivatives in Arabidopsis. Using advanced analytical methods and pathway-impaired mutants, we show that JAO enzymes produce exclusively 11OH-JA and are preferentially recruited in response to fungal infection where 11-O-Glc-JA accumulates as its main glucosylated derivative. Conversly, mechanical wounding triggers the dominant formation of 12-OH-JA (and its derivatives 12-O-Glc-JA and 12-HSO4-JA) as a cleavage product of the JA-Ile catabolite 12OH-JA-Ile by amidohydrolase activity. Our findings elucidate the 11OH-JA biosynthetic pathway and reveal two distinct enzyme systems generating position-specific hydroxy-JA isomers in response to stress. Direct treatment of seedlings with 11OH-JA or 12OH-JA did not trigger specific transcriptional changes, indicating these compounds are inactive derivatives.

Suppression of endothelial ceramide de novo biosynthesis contributes to cardiometabolic diseases

Project description:Metabolic disorders and obesity increase the risk for cardiovascular (CV) disease, including hypertension, atherosclerosis, and myocardial infarction. Systemic endothelial dysfunction, a well-established response to CV risk factors, precedes the development of CV diseases. However, growing evidence suggest that endothelial dysregulation also contributes to metabolic disorders, underlying a centric role of the endothelium in cardiometabolic diseases. The accrual of the sphingolipid ceramide, bioactive molecule, has been causally implicated in endothelial dysfunction in vitro. However, direct in vivo evidence supporting the accrual of ceramide in the endothelium, underlying mechanisms, and pathological implications are lacking. Here, we showed that the suppression rather than the accrual of ceramides is causally linked to endothelial dysfunction, and imparts a pro-inflammatory and pro-atherosclerotic phenotype, contributing to both vascular and metabolic disorders. Mechanistically, the upregulation of Nogo-B and ORMDLs proteins, inhibitors of the first and rate limiting enzyme of the de novo biosynthesis, in the endothelium of obese and diabetic mice suppresses sphingolipid signaling, particularly ceramides and sphingosine-1-phosphate, resulting in vascular and metabolic dysfunctions. Systemic and endothelial specific deletion of Nogo-B restores sphingolipid signaling and functions of the endothelium, improves hypertension and lowers hepatic glucose production. Our results demonstrating that Nogo-B-mediated suppression of sphingolipid metabolism and signaling in the endothelium has pathological implication in cardiometabolic disorders and set a framework for the development of therapeutic strategies to treat these conditions.

Atopic asthma lowers nasal mucosal sphingolipids

Project description:Genetic variations at the 17q21 asthma-risk locus regulate the expression of gasdermin B (GSDMB) and ORMDL3, influencing inflammatory responses and sphingolipid metabolism. While asthma-associated 17q21 variations are known to affect ORMDL3 expression in immune and airway smooth muscle cells, its role in airway epithelial sphingolipid metabolism remains unclear. We investigated whether asthma and 17q21 genetic variations influence sphingolipid composition in the upper respiratory tract and how immune vs. epithelial cells contribute to this process. Sphingolipid profiles were analyzed in nasal fluid and blood from children with and without asthma. We also examined gene expression and sphingolipid composition in nasal epithelial cells and PBMCs from healthy adults homozygous for the rs7216389 C/C and T/T (asthma-risk) genotypes. Children with atopic asthma exhibited lower nasal fluid sphingolipids, including sphinganine, dihydroceramides, and ceramides, independent of corticosteroid use or allergic rhinitis. Asthma was further associated with higher plasma sphingolipids and lower blood cell sphingolipids, the latter mirroring patterns in nasal fluid. In PBMCs, the T allele increased ORMDL3 expression, suppressing de novo sphingolipid synthesis. However, in nasal epithelial cells, the T allele mainly increased GSDMB and there was no effect on sphingolipid metabolism. These findings establish nasal fluid sphingolipid profiling as a potential marker for atopic asthma and provide evidence of a cell-type-specific effect of 17q21 genetic variants. While ORMDL3-mediated sphingolipid suppression occurs in PBMCs and not airway epithelial cells, its systemic effects may contribute to lower airway sphingolipids in asthma.

Chronic Dosed-Feed Toxicity/Carcinogenicity Study of 2-Hydroxy-4-Methoxybenzophenone (HMB, C10260; CAS No. 131-57-7) in Sprague-Dawley rats (G822872B)

Project description:2-Hydroxy-4-methoxybenzophenone (HMB) is an approved Food and Drug Administration (FDA) over-the-counter Category I product considered to be a deterrent to ultraviolet (UV) radiation-induced skin cancer. HMB is used in pharmaceuticals, plastic products, fabric coatings, finishes, and cosmetics. Exposure to HMB occurs through both occupational and consumer routes. HMB is used in skin moisturizing products and sunscreen lotions. In plastics manufacturing, HMB is used in surface coatings and polymers including ABS resins, cellulosic esters, polyesters, polystyrenes, rubber, flexible and rigid vinyl, and vinylidene chloride. HMB readily penetrates the skin and bioaccumulates in lipid-rich tissues. Uptake from the gastrointestinal tract is rapid. This study will characterize the toxicologic and carcinogenic potential of HMB administered in dosed feed to Sprague-Dawley rats given its potential for widespread human exposure as a broad-spectrum sunscreen.

PNPLA1 has a crucial role in skin barrier function by directing acylceramide biosynthesis

Project description:Mutations in PNPLA1 (patatin-like phospholipase domain containing 1) cause autosomal recessive congenital ichthyosis, but the mechanism involved remains unclear. Here we show that PNPLA1, an enzyme expressed in highly differentiated keratinocytes, plays a crucial role in the biosynthesis of acylceramide, a lipid component essential for skin barrier function. Pnpla1-deficient mice showed neonatal lethality due to epidermal permeability barrier defects with severe transepidermal water loss, decreased intercellular lipid lamellae in the stratum corneum, and impaired terminal differentiation of keratinocytes. In Pnpla1–/– epidermis, three unique linoleate-containing lipids, including acylceramides, acylglucosylceramides and (O-acyl)-ω-hydroxy fatty acids, were almost absent with reciprocal increases in their precursors ω-hydroxy (glucosyl)ceramides and ω-hydroxy fatty acids, indicating that PNPLA1 catalyzes the ω-O-esterification step with linoleic acid to form acylceramides. Our results suggest that PNPLA1 represents the missing piece in the process of acylceramide biosynthesis required for establishment of a permeability barrier.

Sac1 interaction proteomics of ALFA Sac1 and mutants

Project description:Sphingolipids are structural membrane components that also function in cellular stress responses. The serine palmitoyl-transferase (SPT) catalyzes the rate limiting step in sphingolipid biogenesis. Its activity is tightly regulated through multiple bind-ing partners, including Tsc3, Orm proteins, ceramides, and the phosphatidylinositol-4-phosphate (PI4P) phosphatase Sac1. The structural organization and regulatory mechanisms of this complex are not yet understood. Here, we report the high-resolution cryo-EM structures of the yeast SPT in complex with Tsc3 and Orm1 (SPOT) as dimers and monomers and a monomeric complex further carrying Sac1 (SPOTS). In all complexes, the tight interaction of the down-stream metabolite ceramide and Orm1 reveals the ceramide dependent inhibition. Additionally, observation of ceramide and ergosterol binding suggests a co-regulation of sphingolipid biogenesis and sterol metabolism within the SPOTS com-plex.

Aging stress and ceramide dependent RNA sequencing expression profiling depends on ceramide synthase 6

Project description:The bioactive sphingolipid ceramide, generated by ceramide synthase, is an adaptive stress effector induced in response to various stress stimuli such as aging. In fact, ceramide synthase (CerS) was first discovered in yeast as a longevity assurance gene (LAG1), as the reduced ceramide generation consequent to LAG1 deletion increased longevity. There are six homologues of mammalian LAG1/ceramide synthases (CerS1-6) that generate ceramides with different fatty acid chain lengths with distinct functions and hydrophobicity. Ceramide can be metabolized by sphingosine kinase 1 or 2 for the generation of pro-survival sphingosine 1-phosphate (S1P), which enhances immune cell egress to the blood stream, activating immune function. Ceramide is known to induce mitophagy and cell death. Here we investigated the role of this sphingolipid pathway in immune function, specifically T cell functions, via high throughput expression profiling.

Lignocellulose fermentation to ethanol or lactate by giant panda gut microbiomes depends ultimately on pH rather than portion of bamboo

Project description:Giant panda are carnivorous bears which feed almost exclusively on plant biomass (i.e. bamboo). The potential contribution of its gut microbiome to lignocellulose degradation has been mostly investigated with cultivation-independent approaches. Recently, we reported on the first lab-scale cultivation of giant panda gut microbiomes and described their actual fermentation capacity. Fermentation of bamboo leaf using green dung resulted in a neutral pH, the main products being ethanol, lactate and H2. Fermentation of bamboo pith using yellow dung resulted in an acidic pH, the main product being lactate. Here, we cultivated giant panda gut microbiomes to test 1) the impact of mixed dung as inoculum; 2) the fermentation capacity of solid lignocellulose as opposed to organics-rich biofluids in the dung; 3) the artificial shift of pH from neutral to acidic on bamboo leaf fermentation. Our results indicate that i) gut microbiomes fermentation of solid lignocellulose contributes up to a maximum of 1/3 even in the presence of organics-rich biofluids; ii) alcohols are an important product of bamboo leaf fermentation at neutral pH; iii) aside hemicellulose, gut microbiomes may degrade plant cell membranes to produce glycerol; iv) pH, rather than portion of bamboo, ultimately determines fermentation profiles and gut microbiome assemblage.

Project description:Patulin, 4-hydroxy-4H-furo[3,2c]pyran-2(6H)-one, is one of the most popular and characterized mycotoxin observed in agricultural products. However the mechnisims of toxicity by this chemical is not well understood. Thus, we characterized the cytotoxicity of patulin using yeast transcriptome systems. The expression profile of mRNA after exposure yeast cells to patulin was similar to those after the treatment by antifungal agricultural chemicals such as thiuram, maneb, and zeneb. The Patulin treatment activated protein degradation especially proteasome activities, sulfur amino acid metabolisms, and oxidative stress defense system. DNA alkylation damages was also suggested and this seems to be repaired by recombinational and excision repair mechanism. We also list the indicator genes for the detection of patulin in agricultura product. Keywords: stress response
