Proteomics analysis of transgenic switchgrass overexpressing melatonin biosynthesis genes

ABSTRACT: To obtain better insight into the underlying mechanism of melatonin in switchgrass growth, flowering and defence, we performed proteomics analyses between transgenic lines overexpressing melatonin biosynthesis genes and transgenic control (expressing only the empty vector) plants.

Induction of secondary cell wall biosynthesis genes and their regulators by melatonin in Glycine max

Project description:Plant lodging severely reduced crop yield and quality. Different plant growth regulators (PGRs) have been applied to improve lodging resistance through the regulation of physiological changes, especially on the increase of stem thickness and strength. Melatonin is a pleiotropic PGR for the regulation of plant growth and development. In this study, we demonstrated that the exogenous treatment of melatonin to Glycine max significantly enhanced plant lateral growth by increasing stem diameter. In addition to the stem thickness, secondary cell wall (SCW) deposition acts as another critical factor for stem rigidity for lodging resistance. To understand whether exogenous treatment of melatonin would regulate SCW biosynthesis genes, we performed transcriptomic analyses on the stems of Glycine max with or without melatonin treatment. Through the differentially-expressed-genes (DEGs) analyses, many SCW biosynthesis genes were found to be regulated by melatonin, including the cellulose, hemicellulose and lignin biosynthesis enzymes. We also found that the two known master regulators, NAC and MYB, of SCW biosynthesis genes were induced under melatonin treatment, which further supported our observation on the differential expression of SCW biosynthesis genes. Our study highlighted the improvement of lodging resistance by the exogenous treatment of melatonin through the increase of plant stem thickness and the regulation of SCW biosynthesis genes and their upstream TFs in Glycine max.

Evaluation of transgenic proteins in transformed Camelina sativa leaves

Project description:The goal of this study was to detect and quantify the presence of transgenic proteins in transformed C. sativa plants. Experimental plants were transformed with recombinant proteins of a synthetic carbon fixation cycle. Plants expressing the full suite of genes of the synthetic cycle are labeled as “T”. Plants expressing only half of the synthetic cycle are labeled as “C”. Plants transformed with an “empty vector” are labeled as “EV” and serve as a negative control. Five biological replicates were included for each treatment. Additionally, one chloroplast enrichment sample was generated for each line using a commercially available chloroplast isolation kit (Sigma Product #: CPISO). Leaf tissue from photosynthetically-active leaves was harvested and immediately flash frozen with liquid nitrogen. This tissue was stored at -80ºC until it was ground to a fine powder under liquid nitrogen. The tissue was then weighed into 200 mg aliquots. Amino acid sequences of the transgenic proteins were used as a reference and “empty vector” control plants were used as a negative control.

OsCOMT, encoding a caffeic acid O-methyltransferase in melatonin biosynthesis, increases grain yield through regulating leaf senescence and vascular patterning

Project description:Melatonin, a natural phytohormone present in most plants, plays multiple roles in plant growth and stress responses. Although melatonin biosynthesis-related genes have been suggested to possess diverse biological functions, their roles and functional mechanisms in regulating rice grain yield remain largely unexplored. Here, we revealed that a rice caffeic acid O-methyltransferase (OsCOMT) gene is involved in melatonin biosynthesis through in vitro and in vivo evidences. Transgenic assays show OsCOMT significantly delays leaf senescence at the grain filling stage, and then improves photosynthesis efficiency. Further experimental and transcriptomic data suggest that OsCOMT inhibits the degradation of chlorophyll and chloroplast, which in turn delay leaf senescence. Histological analysis also reveals the role of OsCOMT in the development of vascular bundle system in rice. The levels of melatonin and cytokinin were significantly increased in the culm of OsCOMT-overexpression plant relative to those of the wild-type (WT). In the OsCOMT-overexpression line, the cytokinin-biosynthesizing genes were up-regulated and the cytokinin-degrading genes were down-regulated, thereby increasing the cytokinin levels compared with the WT. Thus, OsCOMT-mediated vascular patterning may result from the crosstalk between melatonin and cytokinin. More importantly, OsCOMT significantly increased grain number and yield production of rice in various background, including Nipponbare (NIP) and Suken118 (SK118). Our findings show novel insights into melatonin-mediated leaf senescence and vascular patterning, and provide a new strategy to enhance rice yield production.

Melatonin enhances plant growth and abiotic stress tolerance in soybean plants

Project description:Melatonin is a well-known agent that plays multiple roles in animals. Its possible function in plants is less clear. In the present study, we tested the effect of melatonin (N-acetyl-5-methoxytryptamine) on soybean growth and development. Both spraying of leaves and seed-coating with melatonin significantly promoted soybean growth as judged from leaf size and plant height. This enhancement was also observed in soybean production and their fatty acid content. Melatonin increased pod number, seed number and seed weight. However, the 100-seed weight was not influenced by melatonin application. Melatonin also improved soybean tolerance to salt and drought stresses. Transcriptome analysis revealed that melatonin up-regulated the expression of many genes and alleviated the inhibitory effects of salt stress on gene expressions. Further detailed analysis of the affected pathways documents that melatonin likely achieved its promotional roles in soybean through enhancement of genes involved in cell division, photosynthesis, carbohydrate metabolism, fatty acid biosynthesis and ascorbate metabolism. Our results demonstrate that melatonin has significant potential for improving of soybean growth and seed production. Further study should uncover more about the molecular mechanisms of melatoninM-bM-^@M-^Ys function in soybeans and other crops. Four different treatments were chosen, water, salt, 100M-BM-5M melatonin and salt plus 100M-BM-5M melatonin. The comparison of salt/melatonin-treated sample versus water-treated sample reveals salt or melatonin induced transcriptome changes. The comparison of melatonin plus salt treated sample versus salt-treated sample reveals melatonin induced changes when salt exists.

Genes differentially expressed between Bmi1-overexpressing and empty vector control neural stem/progenitor cells
