Project description:Aerobic glycolysis is a hallmark of virus-infected cells, leading to substantial accumulation and secretion of lactate. However, the regulatory roles of lactate during viral infections are still poorly understood. Here, we report that human cytomegalovirus (HCMV) infection leverages lactate to induce widespread protein lactylation and promote viral spread. Lactyllysine decorates intrinsically disordered regions (IDRs) of proteins, regulating viral protein condensates and immune signaling transduction. Dynamic lactylation of immune response pathways, a feature shared during infection with herpes simplex virus 1 (HSV-1), suppresses immunity through regulation of RBM14 and IFI16. K90 lactylation of the viral DNA sensor IFI16 blocks recruitment of the DNA-damage response master kinase DNA-PK, preventing IFI16-driven gene repression and cytokine responses. Together, we characterize global protein lactylation dynamics during virus infection, finding virus-induced lactate contributes to its immune evasion through direct inhibition of immune signaling pathways.

Project description:The novel protein acylation modifications have played a vital role in protein post-translational modifications. However, the functions and effects of the protein acylation modifications in lung adenocarcinoma are still uncertain. Currently, there is also a lack of global identification of acylation modifications in lung adenocarcinoma cells. Therefore, in this study, we detected 10 currently known acylation modifications in lung adenocarcinoma cells by western blot. Interestingly, we found that the abundance of lysine succinylation (Ksu), crotonylation (Kcr) and lactylation (Kla) is likely higher. Immediately, we identified the above three modifications and phosphorylation by global mass spectrometry-based proteomics in lung adenocarcinoma cells. As a result, we got 3110 Kla sites in 1220 lactylated proteins, 16653 Kcr sites in 4137 crotonylated proteins, 4475 Ksu sites in 1221 succinylated proteins, and 15254 phosphorylation sites in 4139 phosphorylated proteins. In conclusion, our results provide a proteome-wide database to study Kla, Kcr and Ksu and phosphorylation in lung adenocarcinoma, and our bioinformatics results provide new insights into the role of acylation modification in lung adenocarcinoma.

Project description:Lysine lactylation (Kla) is a novel posttranslational modification, playing important roles in numerous biological processes, including transcription, metabolism, tumorigenesis and nervous system diseases. Using liquid chromatography-tandem mass spectrometry (LC-MS/MS), researchers have identified lactylation in many human organs and other species, but no research on lactylation in human hippocampi has been reported. In this study, we performed global profiling of lactylation in human hippocampi under normal physiological conditions, and we identified 2579 Class I (localization probabilities > 0.75) lactylated sites in 853 proteins. Bioinformatics analysis showed that lactylated proteins were mainly located in the cytoplasm and involved in metabolic pathways and some neurological diseases. Compared with those reported lactylation databases, 434 lactylated proteins were identified for the first time in this study. Our work expands the database on human lactylation and helps advance the study on lactylation function and regulation under physiological and pathological conditions.

However, how histone lactylation functions under glucose-limited conditions remains unknown. Here, we show that lysine lactylation redistributes to transcription start sites (TSSs) during glucose deprivation, thereby altering biological behaviors in canine hemangiosarcoma (HSA) cells. Glucose deprivation significantly decreased global histone lactylation levels, while lactylation peaks were enriched at TSSs of ATF4‑regulated stress‑response, asparagine-synthesis and immune‑related genes. Stress-response gene expressions were upregulated, and ATF4 polyclonal knockout abrogated this activation. [U-13C]glutamine tracing demonstrated that HSA cells synthesized asparagine from glutamine when glucose was scarce, and asparagine supplementation modestly activated cell proliferation. In HSA patient tissues, H3K18la levels were heterogeneous, and M2-like macrophages preferentially infiltrated tumor regions showing low histone lactylation levels. These findings demonstrate that lysine lactylation regulates transcription that supports tumor cell survival and fosters a pro-tumor microenvironment even under glucose-limited conditions.

Project description:Protein lactylation is a newly discovered posttranslational modification (PTM) and involved in multiple biological processes both in mammalian cells and rice grains. However, the function of lysine lactylation remains unexplored in wheat. In this study, we performed the first comparative proteomes and lysine lactylomes during seed germination of wheat. In total, 8,000 proteins and 927 lactylated sites in 394 proteins were identified at 0 and 12 hour after imbibition (HAI). Functional enrichment analysis showed that glycolysis and the TCA cycle related proteins were significantly enriched, and more differentially lactylated proteins were enriched in up-regulated lactylated proteins at 12 HAI vs 0 HAI through KEGG pathway and protein domain enrichment analysis compared to down-regulated lactylated proteins. Meanwhile, ten particularly preferred amino acids near lactylation sites were found in the embryos of germinated seeds: AA*KlaT, A***KlaD********A, KlaA**T****K, K******A*Kla, K*Kla********K, KlaA******A, Kla*A, KD****Kla, K********Kla and KlaG. These results supplied a comprehensive profile of lysine lactylation of wheat and indicated that protein lysine lactylation played important functions in several biological processes.

Project description:Hypoxia can result in tissue dysfunction, metabolic alterations, and structural damage within the pulmonary tissue, thereby impacting lung ventilation and air exchange. The identification of Hypoxia-inducible factor (Hif) 1α as a pivotal mediator in the inflammatory cascade subsequent to hypoxia induction has been established. However, the mechanism remains elusive. To delve deeper into this phenomenon, we have developed a murine model of sustained hypoxia and utilized nanocarriers for the delivery of lentivirus Hif-1α for knockdown purposes. Our findings suggest that under conditions of sustained hypoxia, knockdown of Hif-1α effectively ameliorated SpO2 levels and attenuated lung injury in our murine model. We observed that Hif-1α-mediated Histone Lactylation was evident in the lungs exposed to sustained hypoxia. Through RNA-seq and ChIP-seq profiling, we determined that upregulation of Hif-1α expression in sustained hypoxic lung tissue is essential for inducing lactylation enrichment of inflammatory response genes. Furthermore, knockdown of Hif-1α returned to normal inflammatory cytokines (e.g. TNF-α, IL-6 and IL-1β). Analysis of plasma metabolites from individuals experiencing restrictive/ obstructive lung disease revealed a significant enrichment of the Warburg effect within the sustained hypoxic group. Thus, our study provides compelling evidence supporting the notion that targeting Hif-1α-mediated histone lactylation may represent a promising therapeutic strategy for managing sustained hypoxia-induced lung injury.
