S-nitrosocysteine proteomics of Schistosoma japonicum collected from infected mice and rats

ABSTRACT: Rats and mice were infected with Schistosoma japonicum, and the worms were collected from infected rats and mice. Worms in rats were exposed to high level of NO, while the worms in mice were not. Then a S-nitrosocysteine proteomics of Schistosoma japonicum collected from infected rats and mice was performed and the data were collected.

Project description:To clarify the profile of in BALF exosome collected from mice infected with influenza virus, we infected 100000 pfu of A/Puerto Rico/8/1934 (PR8) strain. BALF was collected at 24, 48, and 72 hour post infection (hpi). For comparison of the profile of the miRNA in BALF exosome induced by innate immune response, we also intranasally inoculated mice with 50 μg of poly(I:C) and collected BALF at 72 hour post inoculation. We found that some miRNAs were common to both influenza virus infectiona and poly(I:C) inoculation, suggesting that exosomal miRNAs in BALF may function in the innate immune response to virus infection.

Transcriptomic Responses to Prion Disease in Rats

Project description:While prion infections have been extensively characterized in the laboratory mouse, little is known regarding the molecular responses to prions in other rodents. To explore these responses and make comparisons, we generated a prion disease in the laboratory rat by successive passage of mouse RML prions. Here we describe the accumulation of prions and associated pathology in the rat and describe the transcriptional impact throughout prion disease. Comparative transcriptional profiling between laboratory mice and rats suggests that similar molecular processes are unfolding in response to prion infection. At the level of individual transcripts, however, variability exists between mice and rats and many genes deregulated in mouse scrapie are not affected in rats. Notwithstanding these differences, many transcriptome responses are conserved between mice and rats infected with scrapie. Our findings highlight the usefulness of comparative approaches to understanding neurodegeneration and prion diseases in particular. We Adapted RML Mouse Scrapie into Rats and measured the resulting gene expression changes in brain as a result of disease progression. Rats were infected by intracranial inoculation with prion isolates obtained by adaptation of mouse RML scrapie prions into rats. Brain samples were collected from third and fourth passage infected rats and age-matched controls at specified timepoints and gene expression profiles obtained. For each time point, 3 diseased and control brain samples were profiled.

Testis proteome of Trypanosoma cruzi infected mice

Project description:Male CD-1 mice (N=8) were intraperitoneally infected by culture-derived Trypanosoma cruzi trypomastigotes of the Brazil strain. One group (N=4) was treated with an antitrypanosomal drug (nifurtimox) on day 74 post-infection (chronic phase) for 20 days. A group of uninfected mice (N=3) was also included as control. All groups (N=11) were enthanized on day 110 post-infection, testes were collected and subjected to 2D-LC-MS/MS analysis. Label-free quantitation revealed that the testes of T. cruzi-infected mice had an altered proteome pattern relative to treated mice and uninfected controls. The analysis also demonstrated that the left and right testes had different proteomic profiles in infected animals.

Project description:During an infection, bacteria are exposed to the complex host immune response and has to adapt to the changing environment in order to successfully grow within tissues. In the present sturdy we analysed the transcriptome of both host (mouse) and bacteria (Salmonella Typhimurium) at different time points during the infection to gain a better insight into the attack mounted by the host to repel the bacterial infection and how the bacteria adapts to it. For this, mice were infected intravenously with Salmonella Typhimurium and spleens were collected for RNA isolation at specific time points. Transcription profile of infected mice was compared to uninfected controls.

Project description:We artificially infected Sprague Dawley (SD) rats and Kunming (KM) mice with type II Toxoplasma gondii (T. gondii) strain (Prugniaud, Pru) to generate toxoplasmosis, which is a fatal disease mediated by T. gondii invasion in the central nervous system (CNS) with unknown mechanisms. We aimed to explore the mechanism of differential susceptibility of mice and rats to T. gondii infection. Therefore, we established a strategy of isobaric tags for relative and absolute quantitation (iTRAQ) to identify differentially expressed proteins (DEPs) of the rats’ and the mice’s brains compared to the healthy groups.
